Abstract/Summary:
The experiment taking Chinemys reevesiis as the research object,and systematically studied the anesthetic effect and drug metabolism of Telazol on Chinemys reevesiis,and summed up the cases of clinical anaesthesia for pet turtles,to provide reference for the application of zorazepam and telazitamin in the clinical anesthesia of turtles.18 artificially cultured Chinemys reevesiis with a body weight of(700±18)g were randomly divided into 6 groups,injected with Telazol doses of 15,20,25,30 and 35 mg/kg,respectively,and the control group was injected with an equal volume of saline.The behavioral characteristics and adverse reactions during anesthesia process,and the induction time,anesthesia time,recovery time were recorded.The anaesthetic effect of zorazepam and telazidine on Chinese pond turtle results showed that : all experimental turtles had no adverse reactions such as vomiting or convulsions,and there were certain differences between the experimental groups with different injection doses.With the increase of the injection dose,the induction period of anesthesia gradually became shorter,and the anesthesia and recovery period gradually prolonged.When Telazol was injected at a dose of 15 mg/kg,the induction period was significantly higher than other experimental groups(P<0.05),and it took a long time to enter the anesthesia.The duration of anesthesia was significantly lower than other experimental groups(P<0.05).During the anesthesia process,the excitement reaction was intense and the effect was general.When the injection dose was 35 mg/kg,the induction period was the shortest,entered anesthesia quickly,and the anesthesia maintenance time is almost the same,but the recovery period was significantly higher than other experimental groups(P<0.05).Comprehensive analysis of different stages of time and the actual situation of clinical operations,when the injection dose of 25 mg/kg,the induction time,anesthesia time and recovery time were 0.42 h,4.33 h and 4.14 h respectively,and with a short induction period,long duration of anesthesia,rapid recovery and other characteristics,it is more suitable as an administration dose for Chinemys reevesiis in clinic.25 mg/kg was chosen as the injected dose by comparing the anesthetic effect.After administration,1 ml of blood was collected at the venous sinus of the turtle at 0,0.10,0.42,0.50,1,2,4.75,8,12,16,24,48,72 and 96 h respectively,the concentrations of zolazepam and tiletamine in plasma were detected by High Performance Liquid Chromatogram(HPLC),and analyzed with software PKsolver2.0 to research the pharmacokinetic of tiletamine and zolazepam.Experimental results show that the concentration-time curves of zolazepam and tiletamine in plasma all were described by a 2-and 1-compartment model.And the concentration-time equation of zolazepam and tiletamine were C=13.462e-0.223t+2.127e-0.055 t,C=10.619e-0.203t+0.279e-0.047 t,respectively.The major pharmacokinetic parameters were as follows: Cmax=11.695 μg/ml、Tmax=1.060 h and AUC=92.470 μg/ ml·h in zolazepam;Cmax=9.654 μg/ml、Tmax=0.482 h and AUC=56.348 μg/ ml·h in tiletamine.It suggested that blood concentration of zolazepam and tiletamineall reached 8.12 μg/ml and 8.96 μg/ml,respectively,The healthy Chinemys reevesiis into anesthesia,to maintain anesthesia,the blood concentrations of zolazepam and tiletamine Should not be less than 7.18 μg/ml and 4.73 μg/ml.Based on the Aquatic Pet Medical Laboratory of Shanghai Ocean University,Telazol was anesthetized for the turtles who came to require a diagnosis and needed anesthesia.The time of induction,anesthesia and recovery,and the behavioral characteristics and adverse reactions during anesthesia were recorded.Telazol applied to turtle clinical anesthesia cases results showed that the intramuscular injection of the 25 mg/kg dose can make the diseased turtles to enter anesthesia and maintain the time required for the operation.Due to the variety of the species,differences in the individual size and health of the tortoise,the duration of each stage of the anesthesia process will also be vary,Overall,Telazol played an ideal anaesthetic effect throughout the operation and facilitated the operation.
