Compared with China's original Chinese cherry(Prunus pseudocerasus L.),sweet cherry(Prunus avium L.)fruit is larger,more beautiful,more delicious,more resistant to storage and transportation,therefore,sweet cherry are favored by consumers in China.In the the 1870 s,sweet cherry was introduced to China,now has more than 100 years of cultivation history,has the reputation of " the earliest fruit of the spring harvest in the northern region ","diamonds in the fruit" in China.Thanks to huge market demand and rich economic returns,the sweet cherry planting industry in China is developing very quickly.However,at present,the sweet cherries produced in China can barely supply domestic fresh sweet cherry market.We still need to import a large amount of sweet cherries.In the current tide of agricultural transformation and upgrading,the sweet cherry planting and processing industry has undoubtedly become an advantageous option for the development of the local economy,and the development prospects are very considerable.Sweet cherry belongs to the gametophytic self-incompatibility category.Since most of the main cultivars are self-incompatible,the pollination tree must be configured in the production,and artificial pollination is sometimes required.The lack of understanding of the self-incompatibility of sweet cherries can lead to low fruit setting rate or high pollination cost.At the same time,there still have a serious problem with the confusion of the names of sweet cherry varieties in China,which is particularly unfavorable for the promotion of new,excellent and special varieties.Identification of the S genotypes of these species was performed by combining the position of the bands after agarose gel electrophoresis and the results of the sequencing with PCR-amplified products.At the same time,the fruit setting rate of 14 sweet cherry pollination combinations was calculated.The purpose of this study was to provide reference for the allocation of pollination trees in sweet cherry production,and to provide a theoretical basis for the evaluation and utilization of sweet cherry germplasm resources as well as for further research on genetic breeding.The main findings of the experiment are as follows:1.The PCR amplification effect of eight common primer combinations on self-incompatibility S alleles of sweet cherry was significantly different.In contrast,the primer combination BFP93/BFP94 had the best amplification effect,and the amplification of Pru-C2/Pru-C3 R and Pru-C2/Pru-C4 R was worse,BFP73/BFP74 and Pru-C2/Pa-C5 were more Poor,Pcons2p/Pcons2 r,PMT2/Pa-C3 R,Pru-C2/Pa-C5 R are the worst.The amplification results of the specific primers BFP208/BFP209,PaS2p/PaS2 r,PaS3p/PaS3 r,PaS9p/PaS9 r,PaS12p/PaS12 r of S1,S2,S3,S9,S12 genotypes were relatively accurate.The specific primers BFP212/BFP213 of the S5 gene did not amplify electrophoretic bands.The experimental results show that the single primer combination is difficult to achieve the purpose of identifying all S genotypes of sweet cherry,and it needs to be reasonably used according to the characteristics of each primer combination.2.The s genotype of 17 sweet cherry cultivars were: Kaving(S3S9),Lovdia(S1S9),Katalin(S3S12),Lite(S3S12),Qinying 1(S3S9),Summit(S1S2),Black Tartarian(S6S9),Teclowoe(S3S6),Yanhong(S4S12),Bing(S3S4),Longguan(S1S9),Zaodaguo(S3S9),Burlat(S3S9),Tomora(S3S6),Qinying 3(S312),Rainier(S1S4),Van(S1S3).The primers used in this study amplified six S genotypes for these 17 sweet cherry varieties,including S1,S2,S3,S4,S6,S9 and S12,with genotypes S3 and S9 occurring most frequently.3.The fruit set rate of Qinying 1 ×Qinying 1 was 1.1%,and the fruit setting rates of Qin Qinying 1 ×Black Tartarian,Qinying 1 ×Van,Qinying 1 ×Longguan were 18.2%,16.1% and 27.4%.The fruit-setting rate of Sunburst was 46.2%,and the fruit setting rates of Sunburst ×Rainier,Sunburst ×Burlat,Sunburst ×Van were 38.8%,25.0% and 31.2%.The rate of fruit fruit setting rates of Lapins ×Lapins was 28.3 %,Lapins ×Rainier,Rainier ×Lapins were 1.7% and 41.3%.Qinying1 is a self-incompatible cultivar,the self-incompatibility rate of Lapins is significantly lower than Sunburst.The S genotype of sweet cherry varieties had an effect on the fruit setting rate of hybrids,and self-incompatibility cultivars need to refer to the S genotype when they are used as pollination trees...
