This experiment studied the preparation method of wood vinegar-Lactobacillus plantarum synbiotics and the application of wood vinegar-Lactobacillus plantarum synbiotics in weaned piglets by feeding experiment.Test one is the antibacterial experiment in vitro.It mainly determined the antibacterial effect of Lactobacillus plantarum and its metabolites on Escherichia coli,Staphylococcus and Salmonella.In the second test,the preparation processes of the synbiotics were determined.The best culture time was determined by determining the growth curve and pH curve of Lactobacillus plantarum.The differences of OD610 between shaker and static and the effect of initial pH on the growth of Lactobacillus plantarum are determined by the microplate reader.Different proportions of wood vinegar were added to the Lactobacillus plantarum medium to determine the best addition amount of wood vinegar by comparing the difference of OD610.Test three is a feeding experiment,144 piglets?Duroc×Landrace×Large White;32d?with an average of?9.41±0.23?kg were randomly selected and allotted to four treatments with six replications of six pigs per replicate.Dietary treatments were as follows:The first group is the control group?basal diet+100mL MRS?;?2?The seond group is the wood vinegar group?basal diet+0.2%wood vinegar+100mL MRS?;?3?The third group is the Lactobacillus plantarum group?100mL Lactobacillus plantarum liquid?;?4?The fourth group is the synbiotics group?basal diet+100mL wood vinegar-Lactobacillus plantarum synbiotics?.Growth performance,blood biochemical indexes and the fecal flora were determined at the end of this experiment.The results showed that:1.Metabolites of Lactobacillus plantarum are the main inhibitory substances.2.The optimum amount for wood vinegar is 0.2%and the initial pH of the medium was6.5;the suitable condition of culture is 37?for 24h in the static state,the number of viable organism of the synbiotics is 4.58×109CFU/mL and the lactic acid concentration is20.36±0.18g/L?3.Growth performance was promoted on the synbiotics group,wood vinegar group and Lactobacillus plantarum.Synbiotics group significantly increased the ADFI of weaned piglets?P<0.05?;wood vinegar group,lactobacillus plantarum group and synbiotics group all had the trend of increasing the ADG and reducing the F/G?P>0.05?.The diarrhea rate of the wood vinegar group,lactobacillus plantarum group and synbiotics group were extremely decreased?P<0.01?,and synbiotics group was lower than wood vinegar group and Lactobacillus plantarum group?P<0.05?.4.Adding wood vinegar-Lactobacillus plantarum synbiotics obviously increased the apparent digestibility of the CP and CF?P<0.05?,and had a trend of increasing the apparent digestibility of the EE?P>0.05?.5.Wood vinegar-Lactobacillus plantarum synbiotics obviously improved the content of serum ALB in weaned piglets?P<0.05?and had a trend to increase the content of serum TP?P>0.05?;wood vinegar group,Lactobacillus plantarum group and synbiotics group all significantly increased the activity of AKP?P<0.05?and decreased the content of TC and BUN?P<0.05?in the serum of piglets.Synbiotics group serum GPT activity was significantly lower than the control group?P<0.05?,but had no difference on the activity of serum GOT?P>0.05?.6.Adding wood vinegar-Lactobacillus plantarum synbiotics significantly improved the content of GH?P<0.05?;there was no significant difference about the content of INS in each treatment group?P>0.05?.7.Adding wood vinegar-Lactobacillus plantarum synbiotics obviously increased the content of serum IgG and IL-4?P<0.05?in weaned piglets;wood vinegar group,Lactobacillus plantarum group and synbiotics group all had a trend to increase the content of serum IgA in weaned piglets?P>0.05?,there was no difference about the content of IL-6 in each treatment group?P>0.05?.8.Adding wood vinegar-Lactobacillus plantarum synbiotics improved the immunity.Wood vinegar group,Lactobacillus plantarum group and synbiotics group all significantly decreased the concentration of MDA in serum of piglets?P<0.05?.The serum SOD and T-AOC in synbiotics group were obvioustly higher than control group?P<0.05?;wood vinegar group,Lactobacillus plantarum group and synbiotics group also had a higher trend in the content of GSH-PX?P>0.05?.9.Wood vinegar group,Lactobacillus plantarum group and synbiotics group all had a effect on promoting the number of lactobacillus and bifidobacteria and they also had a effect on reducing the number of Escherichia coli and Salmonella.Synbiotics group significantly increased the number of lactobacillus?P<0.05?and obviously reduced the number of Escherichia coli?P<0.05?in the feces of weaning piglets.Conclusion:wood vinegar has a good effect on promoting the growth of Lactobacillus plantarum,and wood vinegar-Lactobacillus plantarum synbiotics can improve the production performance and regulate the blood biochemical index,it also can enhance immunity and antioxidant ability and regulate fecal flora of weaned piglets.
