In recent years,in animal,plant,microorganism and human transcriptional group,a kind of RNA molecule,called circular RNA(circRNA),which is stable in cells and has certain functions.It has been shown that circRNA has a higher abundance and more types in biological cells,and some circRNA acts as miRNA sponge in the cells,thus removing the inhibition of miRNA on its target gene and increasing the expression level of the target gene.It is not obvious what circRNA is in pigs and what their possible functions are.This project seeks to reveal the potential role of circRNA in fat metabolism by comparing the differences in circRNA expression between different tissues of Chinese and foreign pigs.In this experimental study,we selected Luchuan pig and Duroc as test subjects.Basic physiological indexes of two kinds of pigs were determined,and it was proved that there were significant differences in back fat thickness.The liver,fat and muscle tissue samples of the two breeds were then collected for mRNA and circular RNA sequencing and analysis.Each group was sampled by three pigs to construct a mixed sample pool,complete transcription and circRNA sequencing of six samples from two pigs,three tissues,and 101.82 GB Clean Data,with an average of 15.46 GB for each sample,moreover,the base percentage of Q30 is also 85.00%and above;the clean reads of each sample were compared with the specified reference genome,and the efficiency ranged from 97.86%to 99.67%,and 18267 circRNA was identified based on Ciri Software;and 23,198 mRNA were identified by comparison and analysis.Through further analysis,1,714 differentially expressed genes and 190 differential expressions of circular RNA were screened from liver tissues of Luchuan pigs and Duroc,and 1,367 differentially expressed genes and 258 differential expressions of circular RNA was screened from the muscle tissues of Luchuan pigs and Duroc,and and 2,173 differentially expressed genes and 110 differential expressions of circular RNA.Because circRNA regulates gene expression by inhabiting miRNA,we express all of the differences to circRNA for source gene prediction,and to annotate and enrich the source genes,the results showed that it was mainly concentrated on the signaling pathways such as transportation and destabilization,signal transduction,carbohydrate metabolism and lipid metabolism.Then,the genetic analysis of the circRNA of the differentially expressed genes of liver,muscle and fat of Luchuan pig and Duroc pig was followed by QTL screening,locating abdominal fat weight and average backfat differential expression of thickness(QTL related to fat Metabolism)circRNA.They are 3:117261346|117270385,15:131201187|131208307,9:144508195|14473873,14:107870129|107873816,2:20100182|20160570,2:20116028|20160570,and 12:40294951140297612.In addition,the high-throughput sequencing of circRNA(transcription group sequencing)analysis in this study indicates that circRNA plays an important role in the metabolism of pig fat and some functions that may be related,it provides a new way of thinking and an angle for the regulation of fat metabolism in pigs and the study of pork quality characters.
