With the development of social economy,pork has become a common food in people’s daily dining table,and people’s requirements for pork’s quality have gradually improved.The quality of pork is closely related to the content of intramuscular fat.Duroc is a typical lean pig in the market,and has higher rate of lean meat.But its intramuscular fat content is low and its taste is poor.Lu Chuan pig is an obese pig breed with richer intramuscular fat and good taste.LncRNA used to be the "noise" of transcription,but recent research results show that lncRNA has functions in various life activities,and many IncRNAs have been found to play an important role in fat metabolism.Therefore,it is of great significance to search for lncRNAs related to fat metabolism and explore their targets and regulatory mechanism,which will provide a new way to regulate fat metabolism.Muscle,fat and liver are the three major functional tissues of mammals.However,liver plays an important role in de novo lipogenesis,TG storage and fatty acid synthesis,especially in lipid metabolism.In this study,we collected the liver samples from obese and lean pig breeds for high-throughput sequencing,and detected the differences of IncRNA in the liver of two pig breeds.We used multiple criteria for the original data to screen the differential lncRNA related to fat metabolism,and further explored its association with phenotypic changes and the specific mechanism.In this study,we screened a lncRNA closely related to lipid metabolism.Phenotypic studies showed that it could induce changes in triglycerides and mitochondria in cells,and could change the expression of genes related to lipid metabolism pathway.Among them,the expression of MTP mRNA,which is related to lipid transport,had the most significant difference,but did not change the expression of MTP protein.Overall,through bioinformatics analysis and experimental validation,we found a new lncRNA related to fat metabolism,and explored its tissue distribution and function,confirmed its regulatory role on fat metabolism,but its targets and mechanism need further study and exploration.
