In order to explore the polyploidy mutagenesis and identification techniques of Atractylodes chinensis(DC.)Koidz,a new autotetraploid germplasm of A.(DC.)Koidz was obtained.In this study,diploid A.(DC.)Koidz seeds were used as materials,soaked germinating seeds in 0.10% colchicine plus 2.00% DMSO with seed soaking method for 3h,6h,12 h and 24 h.Soaked dry seeds in 0.05% colchicine plus 2.00%DMSO for 36 h,48h and 60 h.According to soma identification and flow cytometry identification,confirmed ploidy level of mutagenesis plant.At the same time,compared the plant configuration feature,micro-character and root and stem characteristics of mutagenesis and diploid A.(DC.)Koidz.The results showed that different concentration of colchicine with different treatment time and different seeds treatment state,the better induce effect is germinating seeds mutagenize for 6h,with1.67% inductivity and 5 strains of autotetraploid.This study has been obtained 11 mutant plants in all,which including 9 strains of autotetraploid and 2 strains of chimaera.Compared with the diploid control plants,the A.(DC.)Koidz mutant plants showed different degrees of leaf-shaped rounding,thicker leaf folds,and deeper leaf color.The number of stomata per unit area of the mutant plants was significantly higher than that of the diploid control plants.Reduce,the length and width of the pores increase.This study has been laid foundation for A.(DC.)Koidz germplasm innovation and new varieties selection.
