The application of goat frozen semen is conducive to improving the utilization of breeding goat.However,due to the high sensitivity of goat sperm to low temperature,the vitality of frozen-thawed goat sperm is low and the sperm structure is seriously damaged.Therefore,screening the antifreeze substances for goat semen cryopreservation is of great significance.The traditional cryoprotectant is mainly egg yolk(EY),in which low density lipoprotein(LDL)play a major role in antifreeze protection.This experiment compared the effects of low density lipoprotein(LDL)and soybean lecithin(SL)on semen cryopreservation of Inner Mongolia cashmere goats.The experimental resμl ts are as follows:1.15% EY(v/v)as the control group,the effects of adding 0.07g/ml,0.09g/ml,0.11g/ml and 0.13g/ml LDL to the frozen diluent on the cryopreservation of cashmere goat semen were compared.After semen frozen and thawed,0.09g/ml LDL significantly improved the sperm motility parameters,structural integrity and mitochondrial membrane potential compared with the control group(p<0.05).2.15% EY(v/v)as the control group I and 1.5% SL(sigma,m/v)as the control group II,the concentrations of 1%,1.5%,2%,2.5% and 3% SL(Coolaber,m/v)in the frozen diluent on the cryopreservation of cashmere goat semen were compared.After semen frozen and thawed,the sperm linear motion,structural integrity and mitochondrial membrane potential of 1.5% SL(cooler)group were significantly higher than those of control group I and control group II(p<0.05).The effect of SL-Coolaber is better than SL-Sigma.3.With 15% EY(V/V)added as the control group,the freezing preservation effect of cashmere goat semen with LDL and SL Coolaber(0.02g/ml LDL+1% SL Coolaber,0.02g/ml LDL+1.5% SL Coolaber,0.02g/ml LDL+2% SL Coolaber,0.04g/ml LDL+1% SL Coolaber,0.04g/ml LDL+1.5% SL Coolaber,0.04g/ml LDL+2% SL Coolaber)added in goat freezing diluent.After semen freezing and thawing,the sperm motility parameters of 0.04g/ml LDL+1.5% SL Coolaber group Plasma membrane integrity rate,acrosome integrity rate and mitochondrial membrane potential were significantly higher than those of the control group(P<0.05),and the thawed sperm indexes of 0.04g/ml LDL+1.5% SL Coolaber group were also better than those of 0.09g/ml LDL group and 1.5% SL(Coolaber)group.In addition,the sperm of 0.04g/ml LDL+1.5% SL Coolaber group survived the longest after thawing,up to 13 hours,indicating that the synergistic protective effect of LDL and SL Coolaber was more conducive to the cryopreservation of goat semen.In conclusion,in the process of semen cryopreservation of Inner Mongolia cashmere goats,the combination of 0.04g/ml LDL and 1.5% SL-Coolaber in the diluent can effectively improve the cryopreservation effect of cashmere goat semen.
