Bovine mastitis is a common complex disease of dairy cows, and there are little research on the epidemic regularity of dairy cow mastitis in Henan Province, the species of main pathogenic bacteria and their drug resistance on the bacteria. This study investigated the incidence of bovine mastitis in Henan Province, the main pathogenic bacteria and their drug resistance, and the changes of serological indexes of mastitis cows. And these provided reference and guidance for the prevention and control of dairy cow mastitis in Henan province. The study is divided into three parts:1.Investigation on the incidence regularity of mastitis in dairy cows in Henan provinceThe total incidence of bovine mastitis in Henan province was 71.67%,which the incidence of subclinical and clinical mastitis was 60.48%and 11.19%respectively;The incidence of mastitis was the highest between November and December, and had a significant difference(p<0.05)between May and June; The incidence of mastitis had a significant difference between the second parity and the first parity(p<0.05);The incidence of mastitis had a significant difference between daily milk production over 25Kg and the daily milk production below 15Kg(p<0.05);The incidence of mastitis had a significant difference between intensive dairy farms and breeding district(p<0.05);The incidence of bovine mastitis had a significant difference between milk machine abnormal operation and normal operation condition(p<0.05).2.Diagnosis and drug resistance of main pathogens from cow mastitis in Henan provinceThrough isothermal nucleic acid amplification microfluidic chip analysis to detection 210 positive milk sample,287 strains of pathogens from mastitis were detected,Streptococcus uberis(41.11%),Corynebacterium bovis(26.13%),Streptococcus agalactiae(11.85%)is main paththogens of bovine mastitis in Henan province. In addition,E.coli(5.57%),M.bovis(5.92%)were secondary paththogens that were infected with mastitis.163 strains pathogenic bacteria were isolated by traditional milk bacteria culture.Confirmed Streptococcus uberis(36.81%),Corynebacterium(20.86%)and Streptococcus agalactiae(19.63%)were popular bacteria of cow mastitis in Henan province as well. There was no significant difference in the detection rate of major bacteria in mastitis between the two methods(p>0.05).Appearently,streptococcus,bacilli were dominant floraof mastitis in Henan province.Drug sensitivity test showed that the four major pathogens of bovine mastitis, include Streptococcus uberis,Streptococcus agalactiae,Corynebacterium and Escherichia coli were highly sensitive to ciprofloxacin, ofloxacin and enrofloxacin, resistant to penicillin, amoxicillin, ciprofloxacin and enrofloxacin were preferred drugs to treatment cow mastitis for dairy farm.3.Study on serum indexes of dairy cow mastitisThe result of comparison of cow serum immune indexes, biochemical indexes and antioxidant indexes between health and mastitis cows showed that the cow serum IL-1(3,IL-8 and ALB had a high significant difference between two groups(p<0.01),the cow serum IL-6 and AST had a significant difference between two groups(p<0.05),the cow serum TNF-?,TP and GLB had no significant difference(p>0.05);the cow serum Ve and Vc had a high significant difference between two groups(p<0.01),the cow serum Fe and Zn had a significant difference between two groups(p<0.05),the cow serum Cu had no significant difference(p>0.05).The results of correlation analysis of serum indexes between health and mastitis groups showed that the cow serum IL-1?,IL-6,IL-8,and TNF-? had a high significant correlation(p<0.01);the cow serum IL-6 and TNF-a had a high significant positive correlation(p<0.01),IL-6 and IL-8 had a significant positive correlation(p<0.05);the cow serum IL-8 and TNF-a had no significant correlation;the cow serum ALT and TP,ALB had a high significant positive correlation(p<0.01);ALT and AST had a significant positive correlation(p<0.05),ALT and GLB had no significant correlation;the cowserum AST and ALB,TP had a significant positive correlation(p<0.01),AST and GLB had a significant positive correlation(p<0.05);the cow serum TP and GLB,ALB had a high significant positive correlation(p<0.01),ALB and GLB had no significant correlation;the cow serum Fe and Vc had a significant positive correlation(p<0.05),Fe and Zn had a high significant correlation(p<0.01);the cow serum Cu and other antioxidant indexes had no significant correlation;the cow serum Ve and other antioxidant indexes had no significant correlation.
