This experiment was divided into two parts.Firstly,determining the activity of SODm and the protective effect of SODm on oxidatively damaged IPEC-J2 cells;Secondly,SODm was added to the broiler diet to study the effects of different levels of SODm on the growth performance,antioxidant capacity and enzyme activity in broilers,and provide theoretical basis for the application of SODm in broiler chicken feed.Experiment 1,the effect of SODm on antioxidant capacity of IPEC-J2 cells.The activity of SODm was measured by NBT method,and the results showed that SODm had SOD activity of 2 200 U·mg-1.The IPEC-J2 cells were divided into 8 groups:control group,H2O2model group,0.5 U·mL-1 SODm+H2O2 group,5 U·mL-11 SODm+H2O2 group,50 U·mL-1SODm+H2O2 group,0.5 U·mL-1 SOD+H2O2 group,5 U·mL-11 SOD+H2O2 group,50 U·mL-1SOD+H2O2 group,3 holes of each group and repeated 2 times.The contents of SOD,GSH-Px,T-AOC and MDA in each group were detected.The results show that:The levels of SOD,GSH-Px and T-AOC in the H2O2 model group were significantly lower than those in the control group,except for the 0.5 U·mL-1 SODm group,the other groups were significantly higher than the model group?P<0.05?.The MDA content in cells of model group was significantly higher than other groups,except the 5 U·mL-1 SOD group,the MDA content in the other groups was significantly higher than that in the control group?P<0.05?,and the 5 U·mL-11 SODm group was significantly higher than 5U·mL-1 SOD group?P<0.05?.This indicates that SODm could increasing the antioxidant capacity of IPEC-J2 cells.Experiment 2:The effect of SODm on growth performance,antioxidant performance and enzyme activity of broilers.The single-factor experimental design was used to select 432 broiler chickens?AA broiler chickens?with similar body 1 day old,divided into 6 groups,with 6 replicates in each group and12 chickens in each replicate.Six SODm levels were set at 0,0.10%,0.15%,0.20%,0.25%,0.30%,respectively.Experimental lasted for 42 days.At 21 and 42 days of age,2 broilers were selected from each repeated for jugular vein blood collection and slaughter,and detected serum immunoglobulin?IgM,IgA,IgG?,antioxidant capacity?the SOD,GSH-Px,T-AOC,MDA in serum and intestinal mucosa?,digestive enzyme activities of duodenum and jejunum?lipase,trypsin,pepsin,amylase?,relative weight and length of digestive tract,content pH and gut microbe.The results are as follows:?1?Different levels of SODm in diet had no significant effect on ADFI,ADG and F/G in broilers?P>0.05?.?2?At 21 days of age,compared with the control group,the SOD activity was significantly lower in the SODm0.30 group?P<0.05?,and the content of T-AOC in SODm0.10 group was significantly higher than other groups.Liver SOD activity was significantly higher than the control group?P<0.05?.At 42 days of age,the SOD activity of SODm group was significantly higher than that of the control group,and the value of SODm0.20 group was the highest?P<0.05?.The MDA content in serum of SODm0.15 group significantly decreased?P<0.05?.The activity of SOD in SODm0.15 and SODm0.25 groups was significantly higher than that in the control group?P<0.05?.The SOD activity in SODm0.10,SODm0.15,SODm0.20 and SODm0.25 groups was significantly higher than that in the control group?P<0.05?.?3?At 21 days of age,the addition of SODm0.20 significantly increased SOD and GSH-Px activity in duodenal mucosa and decreased MDA content?P<0.05?.SODm0.15 and SODm0.20 group jejunal mucosa GSH-Px activity was significantly higher than SODm0.25 group?P<0.05?;the addition of SODm could significantly decreased MDA content?P<0.05?.At 42 days of age,compared with the control group,the level of T-AOC in the duodenal mucosa of the SODm0.30group was significantly increased?P<0.05?,and the content of MDA in duodenal mucosa was significantly decreased apart from SODm0.10.The SOD activity of the jejunum mucosa was significantly increased in the SODm0.10,SODm0.15 and SODm0.20 groups.The level of T-AOC in jejunum mucosa was significantly increased?P<0.05?apart from SODm0.10.The content of MDA in jejunum mucosa of SODm0.20 group was significantly lower than that of the control group?P<0.05?.?4?At 21 days of age,duodenal pepsin activity in SODm0.15 and SODm0.20 groups was significantly higher than other groups?P<0.05?;duodenal trypsin was significantly higher in SODm0.10 and SODm0.20 groups than other groups?P<0.05?;the activity of jejunum amylase in SODm0.10 group was significantly higher than that in other groups except SODm0.30 group?P<0.05?.At 42 days of age,the activity of duodenal pepsin in SODm0.20 group was significantly higher than that in other groups except SODm0.15?P<0.05?.In summary,based on the analysis of the above indicators,SODm can increase the antioxidant capacity of IPEC-J2 cells against H2O2-induced oxidative damage.Adding SODm to the diet had no significant effect on the growth performance of broilers;adding 0.14%0.22%SODm could improve the antioxidant capacity and duodenal digestive enzyme activity of broilers.
