Lily is an important commodity flower in the world which has many varieties.However,our country is very backward in the breeding of lily.At present,the varieties used in cut flowers,potted flower production and garden are basically from foreign varieties.In addition,the production technology of lily bulbs in China is backward,which causes more than 90%of the bulb of cut flower production to be imported,which leads to high production cost,which seriously restricts the development of lily industry in China.Therefore,on the basis of the early work of our group,a study was carried out on the propagation technology of the Asiatic hybrids new FO varieties 'Fendai' and 'Huigui' which were selected from distant hybridization cross L.x formolongi to the Oriental hybrids 'Sorbonne'.The effects of storage time and bulb size on the growth and flowering were studied systematically and the physiological changes during storage were also measured.This paper has an important practical value for the cultivation and application of new varieties of lily,and provides a basis for the study of the bulb production of the new FO species.The main research results are as follows:1.The suitable medium for inducing differentiation and proliferation of 94 hybrids and'Fendai' was studied by using floral organs and bulblet.Here are results:?1?flower organ differentiation of Asiatic Hybrids ranged from strong to weak:pedicel>petals>ovary>style>filaments,and gained 35 hybrids in vitro.?2?Study on 10 of the 35 hybrids:The suitable MS for the Asiatic Hybrids No.2,3,5 and 10 was MS+sugar 30 g·L-1.For the hybrid No.1,4,6 and 9 was MS+6-BA1.0mg·L-1+NAA0.2 mg·L-1+sugar 30 g·L-1.For the hybrid No.7 was MS+6-BA1.5 mg·L-1+NAA0.4 mg·L-1+sugar 30 g·L-1.And the MS+sugar 30 g·L-1 can induce bud,yet MS+6-BA1.0 mg·L-1+NAA0.2 mg·L-1+sugar 30 g·L-1 can induce callus.?3?The suitable MS for the hybrid No.6 and No.9 callus differentiation were 6-BA 1.0 mg·L-1+NAA0.05 mg·L-1 which bud differentiation rate was 75%and 6-BA1.0 mg·L-1+ NAA0.1 mg·L-1which bud differentiation rate was 68.2%.?4?The suitable MS for the propagation of adventitious buds of 'Fendai' scale was MS+ 6-BA1.5 mg·L-1+NAA0.2 mg·L-1.2.The cutting propagation of 'Fendai' and 'Huigui' was regulated by different growth regulators showed that:the optimum IBA concentration for outer scales of 'Huigui' was 50mg·L-1 and 2,4 D concentration was 50mg·L-1.The reproductive effect of the middle scales was stronger than that of the outer scales,while the outer scales were not sensitive to IBA and 2,4 D.The optimum IBA concentration for outer scales of 'Fendai' was 100mg·L-1 and 2,4 D concentration was 50mg·L-1.3.By measuring the growth and development indexes of different bulb size 3-5cm,6-8cm,9-11cm and 12-14cm which were treating in the 4? for different weeks,we know that:the circumference of 3-5cm 'Fendai' and 'Huigui' bulbs did not form flower buds in the year which were cold storaged or not,but low temperature can increase the flowering rate.The larger the bulb size,the greater the effect of low temperature storage on its growth and flowering,which can shorten the time from planting to flowering,and increase the number of flowers.Therefore,12-14cm bulb size should be selected as a commodity bulb production.4.By measuring the growth cone morphological,physiological and growth index of the plant after the cultivation of the 'Fendai' and 'Huigui' which ball size were 12-14cm under the 0-8 week 4? cold storage showed that:The content of IAA/ABA in cold storage for 4 weeks in'Fendai' and 6 weeks in 'Huigui' changed largely.With the prolongation of cold storage time,the content of starch decreased gradually and the content of soluble sugar increased.The content of soluble sugar reached the maximum at 6 weeks for 'Fendai' in cold storage and 7 weeks for'Huigui' in cold storage.At this time,the distance between the top of the bud and the tip of the bud was less than 1cm,and the bulb had broke dormancy.Cold storage can significantly shorten the flowering time of new varieties,increase the number of flowers and prolong the whole flowering period.In the 8 week cold storage of 'Fendai' the flowering time decreased from 158d to 131d,the average flower number increased from 1.43 to 3,the whole flowering period was extended from 12d to 22d.'Huigui' to cold storage for 8 weeks,flowering time decreased from 150d to 137d,the average number of flowers increased from 1 to 2,and the total flowering period increased from 6d to 17d.
