Effect Of Magneyic Treatments On Soil Biological Properities And Soil Microbial Diversity In Rhizosphere Of Pea(Pisum Sativum L.)

The growth of pea can be promoted under magnetic condition,to explore the mechanism of growth enhanced by magnetic treatment,soil biological properties and microbial diversity in rhizosphere of pea were analyzed.Pot experiment was conducted following the next five treatments.i.e.(1)Conventional cultivation(CK);(2)addition with 5 g iron powder(A);(3)addition with 5g magnetized iron powder;(4)addition with 0.2 T magnet+ 5 g magnetized iron powder(C);(5)0.2T magnet only(D).Based on the traditional methods and high-throughput sequencing techniques,soil biological properties and microbial diversity in rhizosphere of pea were analyzed..The main results are as follows:(1)The amount of culturable bacteria and Fungi in rhizosphere soil of pea under 0.2 T magnet+ 5 g magnetized iron powder treatment,were significantly increased than those in control(CK).In addition,compared to the cultivable fungi or actinomycetes,the cultivable bacteria in rhizosphere soil were more significantly responsed to magnetic treatment.Meanwhile,the amount of cultivable actinomycetes was not significantly responsed to magnetic treatment.By contrast,the amount of cultivable microorganisms was significantly decreased than those in control(CK)under A and B treatments.(2)Compared with the control(CK),the microbial biomass carbon,nitrogen and phosphours under the C treatment with 0.2 T magnet +5 g magnetized iron powder were increased 34.0%,46.0%and 44.2%,respectively.In addition,soil enzyme activities,which related to C,N and P cycle werealso improved with 24.6%,23.7%and 31.8%,respectively.On the contrary,soil microbial biomass andenzyme activities were all declined under the A and B treatments.It indicated that the growth of pea would be promoted by continuous and stead magnetic field with increasing the surface area of interaction spot,(3)The highest indexes of bacterial diversity and richness,such as ACE,Chao1,Shannon,etc.,were all obtained in C treatment among the five treatments.Meanwhile,the classification of bacteria at phylum,class,order,family,genus,species and OTU level in C treatment,which obtained the numbers with 36,88,183 344,655,1244 and 3034,respectively.In addition,the Proteobacteria,Actinobacteria,Chloroflexi,Acidobacteria,Firmicutesare the dominant bacterial community at phylum level under different treatments;and the Anaerolineaceae,Microbispora,Acidobacteria,Saccharibacteria,Streptomyces,Bacillus,Actinomadura,Roseiflexus are the dominant population at genus level under different treatments.(4)The highest indexes of fungi diversity and richness,such as Ace,Chao1 and Shannon,were all obtained in C treatment amount the five treatments.Meanwhile,the classification of fungi at phylum,class,order,family,genus,species and OTU level in C treatment,which obtained the numbers with 4,18,4282,132,179 and 261,respectively.In addition,the Ascomycota,Basidiomycota,Zygomycota and k_Fungi are the dominant fungous community at phylum level under different treatments.And the f_Davidiellaceae,Cladosporium,k_Fungi,Fusarium,Gibberella and Alternaria are the dominant population at genus level under different treatments.In summary,the biological indicators of soil fertility and health,such as soil cultivable microorganisms,microbial biomass,enzyme activities and microbial diversity were all significantly improved under C treatment,which used with the 0.2 T magnet + 5 g magnetized iron powder.It indicated that the growth of pea could be promoted by C treatment for accumulating the beneficial microorganisms under higher microbial diversity.
