Research On The Prevention And Treatment Of Osteoporosis In Rats And Pigs With Chinese Herbal Compound

To explore the preventive and therapeutic effects and mechanism of traditional Chinese herbal compound on experimental osteoporosis in rats,and on the basis of this study,the preventive effect of this Chinese herbal compound on osteoporosis in adult aged sows was explored.Medthods:102 3-month-old SPF SD female rats were used to study the prevention and treatment effects of Chinese herbal compound.The prevention trial group was randomly divided into blank group,model group I,A₁?Chinese herbal compound A?,B₁?Chinese herbal compound B?and C₁?raloxifene hydrochloride C?.The treatment group was randomly divided into blank group,model group?,A₂?Chinese herbal compound A?,B₂?Chinese herbal compound B?and C₂?raloxifene hydrochloride C?.Model group I rats were fed with 80 mg·kg^-1·d^-1 Retinoic acid continuously for 14 days,Model group?rats were fed with 80 mg·kg^-1·d^-1 Retinoic acid continuously for 21 days.In the preventive test,drug intervention groups used Chinese herbal compound or raloxifene hydrochloride while making models with retinoic acid.In the treatment trial,drug intervention groups was perfused with Chinese herbal compound or raloxifene hydrochloride after the model was made.The dosage of Chinese herbal decoction is 9.5 g·kg^-1·d^-1 and the dosage of raloxifene hydrochloride is 6.25 mg·kg^-1·d^-1.Bone mineral density?BMD?and blood biochemical indexes?Ca?P?BALP and TRACP-5b?of rats in blank group,model group I and model group II were measured on day 1 after modeling success.Bone mineral density?BMD?,blood biochemical indexes and bone tissue morphology of rats in each prevention group and treatment group were measured on day 14 and day 28 after modeling successin.35 sows aged 4-5.5 years with body weight?220-300 kg?were randomly divided into 7 groups:blank group,Chinese herbal A groups?A₁?A₂?A₃?,Chinese herbal B groups?B₁?B₂?B₃?,each group have 5 sows.Each Chinese herbal compound ultrafine powder group was given three?high,middle and low?doses to feed sows at the ratio of 0.8%,1.2%,and 1.6%,respectively.Collect blood and test blood biochemical parameters on days 0,11 and 21after 20 days of continuous administration.Result display:Chinese herbal A and Chinese herbal B can effectively reduce the serum calcium,phosphorus,TRACP-5b and BALP levels in rats with retinoic acid induced osteoporosis,increase bone mineral density and improve bone microstructure;1.2%and 1.6%doses of Chinese herbal ultrafine powder preparations can effectively increase the levels of BALP and PINP which in pig serum,and reduce the levels of TRACP-5b and CTX-I.The0.8%?1.2%and 1.6%doses of Chinese herbal ultrafine powder preparations can effectively increase the levels of BALP and PINP,and reduce the levels of TRACP-5b and CTX-I.Conclusions:It was proved that Chinese herbal compound A and B could promote the formation of osteogenesis,inhibit bone resorption,regulate the process of bone reconstruction,and increase bone mass and strengthen bone quality in rats and sows.They can improve the level of bone remodeling related enzymes in the serum of sows and regulate the bone metabolism.The clinical application effect is good and worth popularizing and applying.
