In order to improve the clinical curative effect of chinese herbal compound“BingDu Ke”, expansion clinical application range and research the anti-Swine Fever Virusmechanism of action for traditional chinese medicine“Bing Du Ke”. In order to providethe scientific basis for chinese herbal compound formulations reform and industrializedproduction.This study:①Adopting orthogonal design with single factor,then chooseingthe extractum weight of water extraction as index for checking and choosing L9(34)orthogonal table design test to optimize four factors of water extract: time of soaking,water consumption, decoction time, decocting number.②Takeing the Hog choleralapinized virus “C strain” as mode virus, GAPDH and ACTB as referencegene,fluorescence quantitative PCR as testing method, adjusted value of Hog choleralapinized virus as measurement indicators, to research the propagation change of spleenand mesenteric lymph node virus acted on Chinese Herbal Compound after the rabbitswere infected with HCLV.③Takeing the Hog cholera lapinized virus “C strain” asmode virus, ELISA as testing method, IFN-γ concentration as measurement indicators,to research the dynamic change of IFN-γ in rabbit HCLV model acted on antiviralChinese Herbal Compound after the rabbits were infected with HCLV.④Takeing theHog cholera lapinized virus “C strain” as mode virus,body temperature as measurementindicators,to research the effect of Chinese Herbal Compound on the response of regular type Class Swine Fever after the rabbits were infected with HCLV. The studyresults indicate:①The best technology of water extraction is mixing cut crude drug,soaking half-hour, adding12times water into chinese medical materials, decocting3times and1hour each time,the yield of water extraction extractum is57.2%whichclose to the maximum result in the results of orthogonal design.②I n spleen, the lowdose group(2.5g/kg)was significantly higher than control group and high dose group(10g/kg)for Virus content in72hour after the rabbits were injected virus(P<0.05),control group was significantly higher than other three groups for virus content in144hou(rP<0.05).In mesenteric lymph nodes, control group was significantly higher thanlow dose group and high dose group for virus content in72hou(rP<0.05).③The midstdose group(5g/kg)and high dose group can promote rabbit to produce IFN-γ, andwere significantly higher than control group and low dose group(P<0.05)in48hourafter continuation giveing medicine five times(P<0.05), and were very significantlyhigher than control group and low dose group in72hour(P<0.01). Not only the highdose Chinese medicine can induce healthy rabbit to produce IFN-γ,but also it is moreapparent (P <0.01) when the HCLV caused IFN-γ content downing.④The ChineseHerbal Compound can short incubation period of regular type Class SwineFever,prolong the times of enecia,promote temperature to drop.The water extraction process optimization results combine industrial productioncharacteristics for Chinese Herbal Compound “Bing Du Ke”, the best craft combination:soaking half-hour, adding12times water into chinese medical materials, decocting3times and1hour each time,the yield of water extraction extractum is57.2%whichclose to the maximum result in the results of orthogonal design. Poured megadose thefluid into rabbit stomach repeatedly which can signally inhiit proliferation of HogCholera Lapinized Virus“C strain”in the lymphonodimesosteniales and has certaininhibition for proliferation of HCLV in the spleen. Poured megadose the fluid intorabbit stomach repeatedly which can induction product rabbit IFN-γ obviously,especially the high-dosage Chinese medicine not only can induce normal rabbit toproduce IFN-γ,this result will show very outstanding under the circumstance which theHCLV has caused IFN-γ content downing. After injected HCLV72hours,the Chinese Herbal Compound has the effect for promoting temperature to drop andmakeing the body temperatue to perfectly normal for rabbit HCLV model.
