In this paper,the effects of different hormone combinations on callus induction,adventitious bud induction,rooting culture and low temperature stratified seed induction of adventitious buds of Primus mongolica Maxim were studied.The results show that:?1?Mature embryos were used as explants.The best disinfection method was 0.1%HgCI 2 10 min+2%NaCIO 6 min.The infection rate was 5%and the induction rate was 87.67%.?2?In callus induction from mature embryos of Prunus mongolica Maxim,the induction rate was 86.67%when MS+2,4-D 0.7 mg·L-1,+6-BA 1.5 mg·L-1+30g·L-1 was cultured in dark,and 86.87%when MS+2,4-D 0.7 mg·L-1+6-BA 1.5 mg·L-1+sucrose 30 g·L-1+agar 6g·L1 was cultured in dark.?3?In the primary culture of mature embryos of Prunus mongolica Maxim,the differentiation rate of adventitious buds was 83.67%when MS+6-BA 2.0 mg·L-1+NAA 0.2 mg·L-1,sucrose 30 g·L-1+agar 6 g·L-1,and the proliferation rate of adventitious buds was the highest when MS+6-BA 1.5 mg·L-1+NAA 0.2 mg·L-1 sucrose 30 g·L-1+agar 6 mg·L-1.?4?In adventitious bud rooting cultlure of Prunus mongolica Maxim,the rooting rate was 73.80%when 1/2MS?IBA 0.7 mg·L-1+sucrose 30 g·L-1+agar 5.5 g·L-1,and the average number of roots was 3.58.?5?In the germination experiment of Prunus mongolica Maxim seeds,the induction rate reached 72.22%when 1/2MS+IBA 1.0 mg·L-1+NAA 0.2 mg·L-1+sucrose 30 g·L-1+agar 5.5 g·L-1 and the buds grew well,the leaves were abundant and the rooting quantity was the highest.?6?Cryogenic stratified seeds are not easy to disinfect and have high infection rate.The induction rate of adventitious buds is 42.02%?49.43%,but they have faster germination speed,higher germination rate and better growth than those without cryogenic stratified seeds.
