| In order to improve the seedling quality of floating seedlings,the seedlings can adapt to the change of fertilization methods in the field,promote the improvement of tobacco leaf quality,and also alleviate the shortage of mud carbon resources.This study changed the original tobacco floating seedling matrix formula.To explore the effect of biochar on the floating seedlings.This experiment was carried out in the artificial climate indoor of the Modern Tobacco Agricultural Science and Technology Park of Xuchang Campus of Henan Agricultural University from September to November 2018.Three different raw materials of biochar were selected and four additive gradients were set to pass the emergence rate and emergence of tobacco seedlings.The effects of biochar on the growth and development of tobacco seedlings were studied in terms of time,agronomic traits,root growth,photosynthesis intensity,protective enzyme activity,and key enzyme expression of carbon and nitrogen metabolism.1 Although biochar reduces the emergence rate of tobacco seedlings and delays the emergence time,the suitable amount of biochar significantly increases the plant height and number of leaves of the tobacco seedlings.The emergence rate of all added biochar treatment was significantly lower than that of CK treatment,and the degree of inhibition increased with the increase of addition amount.The emergence rate of HT4 treatment was the lowest,which was 28.90%lower than that of CK treatment,and the emergence time increased with the addition of biochar.Lengthening;JT4 treatment has the longest seedling time,which is 5.07 days longer than CK treatment.The plant height and number of leaves treated by DTI,DT2,HT1,HT2,JT1 and JT2 were higher than those of CK treatment.Among them,DT1 and DT2 treatments showed the best performance,the plant height increased by 18.37%and 18.28%,and the number of leaves increased by 26.87%.27.28%.2 Biochar significantly increased the biomass accumulation of tobacco seedlings.In the treatment of peanut shell biochar,the dry weight and root fresh weight of aboveground parts treated by HT1 and HT2 were higher than CK treatment and there was significant difference,and HT2 increased the most.The addition of 33.15%,17.85%,24.13%,and 22.91%,respectively,HT4 treatment was significantly lower than CK treatment;in the addition of rice husk biochar treatment,the dry weight and root fresh weight of aboveground parts treated with DT1 and DT2 were higher than CK.There were significant differences in treatment,and the highest DT2 treatment increased by 36.95%,28.57%,34.48%,and 32.35%,respectively.DT4 treatment was significantly lower than CK treatment;in the addition of wheat straw biochar treatment,all indicators of JT1 treatment were slightly higher.CK treatment,but there was no significant difference.The dry weight and root fresh weight of aboveground part treated by JT2 were significantly higher than CK treatment,which increased by 27.17%,10.71%and 17.24%,20.25%,respectively.At the same time,JT4 treatment was all experimental treatment.The lowest fresh weight and fresh root weight of the middle part of the ground were significantly lower than CK treatment,which decreased by 0.33g,0.02g,0.04g and 5.90mg respectively.3 Biochar increased the relative content of chlorophyll in tobacco leaves and promoted light action.In all treatments,the relative contents of chlorophyll in DT4,HT4 and JT4 were significantly lower than those in CK treatment,and HT4 and JT4 were significantly lower than DT4,which were 14.61%,20.03%and 19.23%lower than CK treatment,respectively.Only DT2 treatment was significantly higher than DT2 treatment.CK treatment was 19.77%higher than CK treatment.The relative content of chlorophyll in DT1,JT2 and JT3 treatment was higher than that in CK treatment,but the difference was not significant.The photosynthetic rate(Photosynthetic,Pn),stomatal conductance(Gs)and transpiration rate(Tr)of DT1,DT2,JT1,JT2,HT1 and HT2 treated tobacco seedlings were significantly higher than CK treatment.The highest phase of Pn,Gs and Tr treated by DT2 was increased by 32.38%,48.38%and 34.62%,respectively,and the intercellular C02 concentration(Ci)was decreased by 15.52%;DT3,HT3,JT3 There was no significant difference in the treatment of Pn,Gs,Tr and CK in tobacco seedlings.Pn,Gs and Tr treated with DT3 and JT4 were slightly higher than CK treatment,while HT3 treatment was slightly lower than CK treatment;and there was no significant difference.The Pn,Gs and Tr of DT4,HT4 and JT4 treated tobacco seedlings were significantly lower than CK treatment,and the average was 7.52%,19.35%and 9.15%lower than CK treatment.4 Biochar can effectively promote the growth of roots of tobacco seedlings.The root vigor of the three biochar additions of 1%and 3%was significantly higher than that of CK treatment.The root activity of DT2,HT2 and JT2 treatment was higher,which was 54.54%and 36.36 higher than that of CK treatment.%,27.27%;when the amount of biochar added was 5%,there was no significant difference between the root activity of the three treated tobacco seedlings and CK treatment;when the biochar addition amount was 7%,the root activity of DT4,HT4 and JT4 treated tobacco seedlings was significant.Below CK treatment,they decreased by 18.78%,22.72%,and 19.01%,respectively.In rice husk biochar treatment,DT2 treatment had the best root morphology index,and the total root length,total heel area,total root volume,number of branches and root tip increased significantly by 19.48%,23.83%,30.76%,and 16.51%,respectively.26.56%,HT2 treatment was the best in peanut shell biochar treatment,but only the total root length and total root volume were significantly higher than CK treatment by 11.68%and 20.51%,respectively.There was no significant difference between other indicators and CK treatment;wheat straw In biochar treatment,JT2 treatment performed best,but only the total root length was significantly greater than that of CK treatment,which was 11.07%.Other indicators were not significantly different from CK treatment.The total root length,total heel area,total root volume,average diameter,number of branches,and number of root tips of the three biochar additions of 7%were significantly lower than those of CK treatment.5 Biochar can significantly increase the protective enzyme activity of tobacco seedlings.In rice husk biochar treatment,the activities of SOD,POD and CAT in DT1 treatment and DT2 treatment were higher than those in CK treatment,and there was significant difference.DT2 treatment was the highest,which was 30.39%,35.48%and 16.67%higher than CK treatment.The activity of SOD,POD and CAT were lower than that of CK treatment and there was significant difference.In the treatment of peanut charcoal and wheat straw biochar,the activity of all treated POD,SOD and CAT decreased with the increase of the addition amount.The JT1 treatment was the highest and significantly higher than the CK treatment,which increased by 25.62%,38.26%,and 16.66%,respectively.The HT4 treatment was the lowest and significantly lower than the CK treatment,which was reduced by 13.79%,20.86%,and 12.96%,respectively.6 Biochar can significantly increase the expression of key enzyme genes in carbon and nitrogen metabolism.The expression levels of granule-bound starch synthase(GBSSI)were increased in 1%and 3%of the three biochars.The expression of GBSSI gene in DT1,DT2 and HT1 was higher than that of CK.Significantly increased 47%,89%,72%,DT4 and JT4 treatment was significantly lower than the control treatment,respectively,17%and 23%lower than CK treatment;three biochar additions were 1%and 3%treatment group starch branching enzyme The expression of(SBE)gene was significantly higher than that of CK treatment.The treatments of DT1,DT2 and HT2 were relatively high,which were 31%,39%and 29%higher than CK treatment respectively.The three biochar additions were 1%and 3%.The expression level of sucrose phosphorylase(SPS)gene in tobacco seedlings was significantly higher than that in the control treatment.The higher DT1,DT2 and HT2 were 48%,55%and 32%higher than CK treatment,respectively.The expression levels of glutamine synthetase(GS1-5,GS1-3)genes in the 1%and 3%treated tobacco plants were significantly higher than those in the control treatment.Different biochars had different effects on the expression of GS1-3 gene.Rice husk biochar is superior to the other two biochars,and the expression of GS1-5 gene is slightly different by different biochars;The nitrate reductase(NR)treated with biochar addition of 1%and 3%was higher than CK treatment,and the performance of rice husk biochar treatment was the best.The treatment resolution of DT1 and DT2 was significantly higher than that of CK treatment,which increased by 39%and 53%,when the addition amount was 7%,the expression levels of NR genes in the three biochars were lower than those in CK treatment,and JT4 was the worst,which was 23%lower than CK treatment.Conclusion:Adding biochar can effectively improve the agronomic traits,photosynthesis ability,root growth,antioxidant enzyme activity,and key enzyme expression of carbon and nitrogen metabolism.The rice husk biochar can be effectively promoted by 3%.The growth and development of tobacco seedlings significantly increased the quality of tobacco seedlings. |
