| Paclitaxel is currently the best first-line drug for the treatment of cancer,and it is essential to solve the problem of insufficient supply of raw materials.The dynamics and differences of the biomass and 10-DAB content of the two seedlings were analyzed by HPLC(high performance liquid chromatography)with 2~6a raw container seedlings and bare rooted seedlings of Taxus yunnanensis as materials.Based on the 10-DAB content and the 10-DAB accumulation per plant,some theoretical support was provided for the selection of suitable garden materials for the establishment of medicinal raw material forests of Taxus yunnanensis and the determination of the best harvest time.The following main findings were obtained:(1)The biomass of container seedlings and bare root seedlings increased with time,and the growth period was from March to September every year.The growth in this stage exceeded 70% of the annual growth,82.8% at highest,the growth in autumn and winter had weakened or stopped.The biomass of the shoots in the container seedlings 2~6a was 29.8g,77.0g,244.0g,537.3g,902.8g,respectively.The biomass of the shoots of bare rooted seedlings 2~6a was 23.7g,64.4g,245.7g,534.2g,898.6g,respectively;2a and 3a container seedlings leaf biomass was significantly higher than bare root seedlings(P < 0.05),the difference fluctuated between 14.1%~24.5%,the difference showed a decreasing trend with time;there was a linear relationship between the biomass of the foliage of two seedlings and the month.The empirical formula was a one-dimensional cubic equation,and the fitting relationship was good.The fitting correlation coefficient was above 0.99.(2)The annual variation trend of 10-DAB content in container seedlings and bare root seedlings increased first and then decreased,the highest value appeared in July or September each year,and the lowest value appeared in January or March.The average annual content of container seedling 2 ~6a was 0.906%,0.934%,0.071%,0.030%,and 0.007%,respectively.The annual average content of bare rooted seedlings in 2-6 years was 0.888%,0.941%,0.067%,0.031%,and 0.007%,respectively.The content of 10-DAB in leaves decreased linearly from 3 to 4a,The highest content of 10-DAB in container seedlings was 1.307% and the lowest was 0.005%,the difference between the two was about 260 times.The content of 10-DAB in bare root seedlings was 1.232% and the lowest was 0.005%,which was about 245 times.The highest value appeared in the 9th month of 3a;the monthly and inter-annual content of container seedlings and bare rooted seedlings were not significantly different(P > 0.05);the 10-DAB content of branches and leaves was linear with the month.The formula was a one-dimensional multiple equation,and the fitting relationship was good,and the fitting correlation coefficient reached 0.95 or more.(3)The cumulative amount of 10-DAB in the leaves and the roots of the container seedlings was basically the same as that in the 10-DAB content.The cumulative amount of 10-DAB in the 3a of the seedlings was significantly higher than that in the other years(P < 0.05);the highest cumulative value of container seedlings was 0.938g/strain and the highest cumulative value of bare rooted seedlings was 0.743g/strain in the 9th month of 3a;the total difference of 10-DAB accumulation in the 2~4a of the two seedlings was significant(P < 0.05),the overall difference of 5~6a was not significant(P > 0.05).(4)At present,the market price of dried yew leaves with a 10-DAB content of 0.15% or more is 50-100 yuan/kg.According to the principle of maximizing the cumulative amount of 10-DAB per plant,it is recommended to carry out the foliage picking when the seedlings grow to the 3rd month of September.At this time,the biomass of the leaves of the container seedlings and the bare root seedlings are 71.8g and 60.3g,respectively,according to 6000 strains/acre,the container seedlings are 69kg/mu higher than the bare root seedlings,the price is 60 yuan/kg,and the multi-income is 4140 yuan/mu.After deducting the previous investment of 1108 yuan/mu,the final income is 3032 yuan/mu.It is recommended to use container seedlings as the garden material,and it is advisable to pick the branches and leaves when the seedlings grow until 3a in September,and relatively high yields can be obtained. |
