Rex rabbit is an economic animal with high breeding efficiency.Rex rabbits are susceptible to diarrhea and even death during weaning due to poor digestive system development and relatively low immunity,which seriously affects the benefits of breeding.Jiawei Sijunzi Granule is made by modern pharmacy technology,which include Sijunzi,Astragalus membranaceus,Radix Bupleuri and so on.Previous studies show that Jiawei Sijunzi Granule has good health effects on weaned Rex rabbits,meanwhile it could significantly reduce intestinal diarrhea and mortality of weaned Rex rabbits.In order to explore the intestinal development and mechanism effects of Jiawei Sijunzi Granule on weaned Rex rabbits deeply.The effects of Jiawei Sijunzi Granule on intestinal length,weight,intestinal structure,immune function,intestinal barrier function and main factors of weaned Rex rabbits were studied by means of physiological and pathological sections,enzymelinked immunosorbent assay,immunohistochemistry and RT-PCR.The effect of nutrient absorption will provide experimental basis for the clinical application of Jiawei Sijunzi Granule.160 healthy Sichuan Rex rabbits aged about 45 days were randomly divided into 5 groups.The normal control group was fed with basic diet.The low-dose group,middle-dose group and high-dose group were fed with 0.5%,1% and 2% Jiawei Sijunzi Granule on the basis of basic diet.The positive control group was fed with 0.8% glutamine on the basis of basic diet.The preliminary period is 7 days and the formal period is 30 days.On the fifteenth and thirtieth days,six rabbits were sampled from each group for sample collection and detection.The results were as follows:1.The results of measurement of intestinal length and relative weight of weaned Rex rabbits showed that the intestinal length of weaned Rex rabbits during the experiment was significantly increased by dietary supplementation of 2% Jiawei Sijunzi Granule,and the intestinal length of weaned Rex Rabbits at the 30 th day of the experiment was significantly increased by dietary supplementation of 0.8% glutamine in the diet.The effects of each group on the relative weight of small intestine of Rex rabbits were not obvious during the experiment.2.The results of intestinal tissue slices of weaned Rex rabbits showed that the dietary supplementation of 2% Jiawei Sijunzi Granule could significantly increase the villus height and the ratio of villus to crypt of weaned Rex rabbits at 15 th day of the experiment.The dietary supplementation of 0.8% glutamine could significantly increase the the ratio of villus to crypt of weaned Rex rabbits at 15 th day of the experiment.3.The results of determination of SIgA content in intestinal tract of experimental Rex rabbits showed that the diet supplemented with 2% Jiawei Sijunzi Granule or 0.8% glutamine could significantly increase SIgA content in duodenum of weaned Rex rabbits,and the diet supplemented with 2% Jiawei Sijunzi Decoction granule could significantly increase SIgA content in jejunum of weaned Rex rabbits during the experimental period.The results of immunohistochemistry showed that dietary supplementation of 2% Jiawei Sijunzi Granule or 0.8% glutamine could significantly increase the expression of SIgA protein in jejunum of weaned Rex rabbits during the experiment.4.The study on intestinal barrier function of experimental Rex rabbits showed that 1% Jiawei Sijunzi Granule could significantly increase the expression of Occludin mRNA in duodenum and ileum of weaned Rex Rabbits on the 30 th day of the experiment.Adding 2% Jiawei Sijunzi Granule to the diet could significantly increase the expression of Zo1 mRNA and Occludin mRNA in jejunum of weaned Rex Rabbits on the 15 th day of the experiment,as well as the expression of Zo1 mRNA and Occludin mRNA in duodenum and jejunum on the 30 th day of the experiment.Occludin mRNA expression.Adding 0.8% glutamine to the diet could significantly increase the expression of Zo1 mRNA and Occludin mRNA in duodenum and jejunum of weaned Rex Rabbits on the 15 th day of the experiment,as well as the expression of Zo1,Claudin1 mRNA and Occludin mRNA in duodenum and jejunum of weaned Rex Rabbits on the 30 th day of the experiment.5.The absorption of main nutrients in intestinal tract of experimental Rex rabbits showed that 1% Jiawei Sijunzi Granule could significantly increase the expression of GLUT2 mRNA in jejunum of weaned Rex Rabbits on the 30 th day of the experiment.The expression of GLUT2 mRNA in jejunum and ileum of weaned Rex rabbits was significantly increased by adding 2% Jiawei Sijunzi Decoction granule to the diet.Adding 1% Jiawei Sijunzi Decoction granule to diet significantly increased SGLT1 mRNA expression in duodenum of weaned Rex rabbits during the experiment.The SGLT1 mRNA expression in duodenum and ileum of weaned Rex rabbits was significantly increased by adding 2% Jiawei Sijunzi Granule during the experiment.Dietary glutamine supplementation of 0.8% significantly increased SGLT1 mRNA expression in duodenum of weaned Rex rabbits on the 30 th day of the experiment.In conclusion,the addition of Jiawei Sijunzi Granule to the diet can increase the length of small intestine,intestinal morphology,intestinal immune function and enhance the absorption of glucose in small intestine.Forward,Jiawei Sijunzi Granule promote intestinal development of weaned Rex rabbits,prevent diarrhea,promote the growth and development of Rex rabbits so as to increase the efficiency of breeding.Adding Jiawei Sijunzi Granule to the basic diet was the best at 2%.The research laid a foundation for the application of Jiawei Sijunzi Granule in weaning health care of Rex rabbits.
