Identification Of The Hybrids Of Vitis Davidii Foex Using Morphology And Molecular Markers

Vitis davidii Foex is one of the characteristic wild grape resources of East Asian populations,it is mainly distributed in the southern part of China,It has strong resistance,good agronomic traits,and good economic benefits.In this paper,using morphology and molecular markers identification of the 64 hybrids of Vitis davidii Foex,the purpose is to combine the results of morphological observations and SSR molecular markers,identify authenticity of hybrids,accelerating the efficiency and process of Vitis davidii Foex breeding.The main experiments and the results were as follows:(1)Morphological observations were performed using characteristics such as leaf characteristics,phenophase,and fruit quality: The basis for the judgment is whether there are paternal features in hybrids,it has been preliminarily established that 54 hybrids have the genetic traits and characteristics of the parents.(2)Using SSR molecular markers to identify of hybrids:Efficient method of DNA extraction from hybrids of Vitis davidii Foex,The SSR reaction system was optimized,Using selected SSR primers obtain the results,that showed that 61 individual plants exhibited the specific PCR bands same as their parents.they were identified as the genuine hybrids.(3)Combined with the results morphological analysis in field and SSR molecular markers,61 individual plants are the genuine hybrids,combined the characteristics of their parents,and there are some chartreuse fruits that is different from the color of the parents,It finds the traits separation of fruit between hybrids.Comparison the results of morphological analysis and SSR molecular markers,it finds that SSR molecular identification method is more accurate than morphological observations,The results suggest that SSR markers could be used as effective for the authenticity of the progenies of grape.It can be used as an effective adjunct way to the breeding of wild germplasm resources of vitis.
