vitis davidii Foex has excellent properties as well as great economic potential, but vitis davidii Foex can hardly strike root which seriously restricted its development and application. This experiment mainly includes the construction of in vitro rapid propagation system of vitis davidii Foex's tissue culture, heating test in harwood cuttings of vitis davidii Foex to promote rooting and the researches on the mechanism of vitis davidii Foex's rooting, aiming at solving the serious problem of vitis davidii Foex's rooting, building up its rapid propagation system of high efficiency, efficiently and profitably promoting the development of this kind of vitis davidii Foex.1. The construction of in vitro rapid propagation system of vitis davidii Foex's tissue cultureIn this experiment, the single bud green cuttings from xiangniang-1(Vitis davidii Foex) was used as materials. Results showed that:the green cuttings with single bud with the treatment in lmolˇL-1 HCl for 30 seconds, then disinfected with 0.1% HgCl2 for 8 min, finally inoculated in solid medium with 20 mg/L tetracycline, can obtain the optimal disinfection effect. That is the most optimal disinfection method; The optimum inducing medium on these explants was MS+6-BA 1.0 mg/L+IB A 0.25 mg/L.1/2MS+6-BA1.0mg/L was the most optimal multiplication medium; Compared with the other varieties, Xiangniang-1's rooting ability was weak.2. The effect of harwood cuttings of vitis davidii Foex by heatingThe experimental material is Xiang-zhenzhu and Ziqiu (vitis davidii Foex) Results showed that:after processing the cuttings with different growth regulators IBA and NAA, those treated by IBA's root effect index were significantly higher than those treated by NAA. It indicated that IBA is an effective root growth regulator for vitis davidii Foex's rooting. These two kinds of vitis davidii Foex's rooting ability both are weak in overall, which when treated with IBA750 mg/L (treatment 8) can obtain good rooting effect and treatment 8 was the optimal rooting treatment; Using electric heating temperature-control process, vitis davidii Foex can obtain high rooting rate.3. Rooting mechanism of vitis davidii FoexWe selected Xiang Niang-1 (vitis davidii Foex), Ziqiu (vitis davidii Foex), ruby seedless. Xiahei seedless as the experimental material. Both ruby seedless and Xiahei seedless are easy to take root and these three varieties were studied through two different ways, namely scanning electron microscope and GC-MS. The results showed that the vitis davidii Foex's anatomical structure was dysplasia. esay-rooting variety's structure is complete boundaries between each part and the structure of the internodes is more mature than stem section, the distribution of raphid in the stem section shows obviously difference among different varieties, the raphid quantity of vitis davidii Foex is little and the distribution range is narrow.Those easy-rooting varieties have large quantities of raphids with wide distribution rage and are rich in inclusions than stem section of the same variety. Through GC-MS detection, it was found that vitis davidii Foex contains varieties of inclusions with a large number of special lipid, most of which hardly take root but rich in polyphenol. A peculiar chemical substance (1,6-Octadien-3-ol,3,7-dimethyl-,2-aminobenzoate) is a kind of toxic substance in vitis davidii Foex, that may have influence on the rooting ability to some extent.
