Growing Development And Muscle Growing Development Regulatory Gene POU1F1 Expression In Qianhua Mutton Merino

Qianhua Mutton Merino is a new fine wool sheep that is bred in Jilin Province which guarantees the development of mutton sheep industry.Growing development is the essential character for evaluating the quality of sheep,and the foundation of feeding management plan.For assessing accurately the characteristics of Qianhua Mutton Merino,especially the growing performance,the weight and the body of male and female sheep of Nucleus group from birth to 24 months were measured.At the same time,immunohistochemistry,qRT-PCR and Western Blotwere used to analyze the expression level of the POU1F1(pituitary specifific transcription factorl,POU1F1)which was closely related to growing development in musculi longissimus thoracis,pectorales and semitendinosus.The aim is to verify the gene regulation for growing development of muscle.And screen the genetic marker for early growth,This study is significant for selection of new breed and acceleration of breeding process.The research results are as follows:1.I measured weight height,body length,chest and cannon circumference of male and female sheep that born in 0 month,3 months(weaned),6 months(sexually mature),12 months(body mature),18 months(initial breeding period)and 24 months(adult period).Body size of male and female Qianhua Mutton Merino was found to increase with growth of age.The results showed that the breed had the characteristics of rapid development,and continued to grew after 1.5 years old.And their chest circumference and body length are both high with good shape.2.The results of immunohistochemical test showed that POU1F1 had positive expression in muscle fiber cytoplasm of longissimus dorsi,pectorales and semitendinosus of Qianhua Mutton Merino.It is indicated that POU1F1 can be expressed in 3 different muscle tissues of Qianhua Mutton Merino.3.QRT-PCR and Western Blot were used to study the difference of POU1F1 in mRNA and protein expression of the longissimus dorsi,pectorales and semitendinosus of Qianhua Mutton Merino.The results showed that both mRNA and protein levels of POU1F1 could express in the longissimus dorsi,pectorales and semitendinosus there is no significant difference(P >0.05)in mRNA and protein levels of three muscles.But there was a tendency that semitendinosus > pectorales > longissimus dorsi.4.The expression of POU1F1 in the muscle of Qianhua Mutton Merino showed that POU1F1 could be used as a genetic marker for growing development of mutton sheep,especially for muscle.
