Qianhua Mutton Merino,a new breed of sheep in Jilin province with the production of meat as the main direction and combine the production of more than 66 homogenous fine wool and the hair follicles structure directly affect the wool yield and quality.Same as other sheep breeds,there is a characteristic of cyclical growth,which present a periodical change from anagen to catagen,to telogen in hair follicles of Qianhua Mutton Merino.Hair follicles cyclical growth is regulated by many signaling molecules including BMP,SHH,Wnt,FGFs,and Wnt10 b plays an important role in regulation of hair follicle regeneration and maintain hair follicle cyclical growth[1-2].Therefore,study the structure and change rule of hair follicles,and expression profile of Wnt10 b in hair follicles of Qianhua Mutton Merino in hair follicles cycle are significant for further breeding work and research on molecular mechanism of hair follicle cyclical growth.In this study,the structure and density changes of hair follicles in anagen,catagen,and telogen of Qianhua Mutton Merino was comparatively studied by histological method.The results showed that there were some differences on hair follicles structure and density of Qianhua Mutton Merino in different periods.In anagen,there were pear-shaped or funnel-shaped primary hair follicle and secondary hair follicle with larger diameter,and density granular matrix cells.When entered catagen,matrix cells and dermal papilla became smaller,and hair bulb became more smaller,hair cells decreased,and inner/outer root sheath pinched in telogen.The variation of primary follicle density in each stage was not significant(p>0.05),the secondary follicle density was maximum in anagen,secondly was catagen,and the density was the minimum in telogen,and differences were significant(p<0.05).The S/P value was the maximum in anagen,and was significantly differentin thecatagen and telogen(p<0.05).In addition,the expressions of Wnt10 b gene and protein of skin of Qianhua Mutton Merino in different periods were determined by real time PCR and western blotting in this study.The results showed that Wnt10 b were expressed in anagen(September),catagen(January),and telogen(March),and presented regularity.The expression was maximum in anagen,secondly was catagen,and the expression was the minimum in telogen,and the determined results of gene and protein were same.By immunohistochemistry,we found that the matrix cells of anagen hair follicle strongly expressed Wnt10 b,and catagen and telogen hair follicle weakly.This implies that Wnt10 b is temporally and spatially related with hair follicle cycle and multiplication of matrix cells.In conclusion,the structure and density of hair follicles of Qianhua Mutton Merino in the hair follicle development cyclewere different.Wnt10 bhave a cyclically changed regular,the highest expression in anagen,and decreased expression in catagen andtelogen.This implies that Wnt10 b may regulate the hair follicle cyclical growth of Qianhua Mutton Merino.
