Reproductive traits in sheep is one of the important economic traits, The reproductive traits of rams are directly related to sheep testicles. Vascular endothelial growth factor(VEGF) is known to induce endothelial cell proliferation, to promote cell migration and to inhibit apoptosis, thus playing a central role in angiogenesis and in the regulation of vasculogenesis.VEGF is one of the most potent stimulus to angiogenesis and vascular endothelial growth factor that have been found.VEGF exerts its biological function by its receptor mediating and may be involved in regulating angiogenesis and vascular permeability of the various organs of the reproductive system. This study analysis of differentially expressed VEGF between sexually immature and mature testes of sheep. The internal organization of the mature testis revealed the dynamic process from the angle of angiogenesis of testis. The purpose of the study was to explore the mechanism of VEGF in the development of testis and spermatogenesis, and to provide a theoretical basis for early use and regulation of the reproductive function.1.This experiment using RT-PCR method analysis of differentially expressed VEGF of Qianhua Mutton Merino sheep core group in 2 months and 12 months old ram testis VEGF mRNA expression differences were detected. The results showed that: MRNA gene VEGF was expressed in testis before and after sexual maturation, but there was a certain difference in expression level. The relative expression of VEGF gene in the testis of December was significantly higher than that in February. The results indicated that VEGF may play an important role in the formation of micro blood vessels of the sheep testis and participate in the growth and development of testis and spermatogenesis.2.Using SP immunohistochemical analysis of VEGF in the tissues of testis before and after sexual maturation. The results showed that: VEGF was expressed in testis before and after sexual maturation. VEGF expression in the early stage of sexual maturation in the primary cells, primary and Sertoli cells, Leydig cells, and cells. VEGF is expressed in all levels of germ cell, Sertoli cell and Leydig cell in the mature stage of sexual maturity. Show that, VEGF by autocrine or paracrine effect of the microenvironment in the testis, spermatogenesis to provide suitable environment. VEGF expression in all levels of sperm that sperm itself is also the target cells of VEGF, VEGF directly involved in spermatogenesis.3. Blot VEGF method was used to detect the difference of western in the testis of sheep before and after sexual maturity. The results showed that the expression of VEGF was significantly higher in December than that in February(P < 0.05). VEGF in testis development and spermatogenesis not only can regulate testicular microcirculation,but also can autocrine or paracrine effects of germ cell proliferation and differentiation and spermatogenesis.
