This paper mainly discusses the key techniques of tissue culture of red leaf begonia,mainly studying the extraction time,disinfection time,type and size of explants,hormone ratio to the induction of explants,adventitious bud proliferation and rooting seedlings,The relationship between the method of refining seedlings and the survival rate of seedlings has established a complete set of red leaf begonia plant tissue culture system,which provides technical support for future development and industrial production.The main research results are as follows:⑴In the acquisition and induction of red leaf begonia explants,disinfectants such as 75%ethanol and 2%NaClO,20%H2O2,and 0.1%HgCl2 disinfectants commonly used in tissue culture are selected for disinfection and sterilization tests.The overall effect of the three disinfectants is 2%NaClO>20%H2O2>0.1%HgCl2.The best method for disinfection of red leaf begonia explants is:75%ethanol disinfection 10s+2%NaClO disinfection 14min(add two drops of Tween).In the study of sampling time,the pollution rate and browning rate are negatively correlated from April to October,so it is concluded that the pollution rate and browning rate are low,and the survival rate is only April,May,and June.During this season,the red leaf begonia explants are better seasoned.In the induction of red leaf begonia explants with different concentrations of plant hormones,the optimal medium ratio was MS+6-BA1.0mg/L+NAA0.3 mg/L,the germination rate reached 85%,callus and the growth of differentiated buds is more.In the study of the effect of the type and length of explants on the induction of red leaf begonia explants,the induction rate of the semi-lignified stem segments was the best among the explant types,and the semi-wood length of 1.5 cm was selected in the tissue culture of the stem segments The induction rate is high when the stem segments are transformed,and the growth potential is good.⑵In the subsequent propagation of red leaf begonia,the optimal ratio of plant hormones at different concentrations is MS+6-BA1.5 mg/L+IBA0.05 mg/L.The buds differentiated in this medium have strong growth potential and rapid proliferation Some,the germination rate is as high as 60%.The optimal ratio of AC to different concentrations is 1.0 g/L,which helps to promote the proliferation of buds.The optimal ratio of GA3 at different concentrations is 1.0mg/L,which is helpful for the growth of seedlings.(3)In the rooting culture of red leaf begonia,the optimal ratio of the combination of basic medium and plant hormone concentration is 1/2MS+IBA0.5 mg/L.When inoculated on the basis of this medium,the seedlings showed excellent growth potential and more than two roots.The study of AC concentration on rooting seedlings showed that 1.0 g/L was the best,with thick seedlings and many roots.⑷In the study of transplanting red leaf begonia tissue culture seedlings,3d treatment without opening the bottle mouth natural light+3d indoor opening bottle opening is the best method for smelting seedlings for transplanting red leaf begonia tissue culture seedlings.Among the effects of different coverings on transplanting of tissue culture seedlings,the survival rate of transplanting when covered with glass beakers was as high as 76.67%.In the study of different transplanting substrates,the substrate with humus+perlite(1:1)ratio is the best cultivation substrate.
