| Plants of the Genus Cerasus is the most iridescent one of ornamental plant in early spring,as the need of seedlings increasing,the request of the rapid cultivation also raising.Carrying out a study on the tissue culture technology of the Genus Cerasus is of great significance that to improve its seedling efficiency for the rapid cultivation of the Genus Cerasus seedlings and enrich the plant-resource of gardening.For these needs,three plants of the Genus Cerasus which has high value of ornamental had been slected in this study:C.yedoensis ‘Somei-yoshino’ has been widely used in afforest and landscape architecture because of its high value of ornamental.The demands of market is large.C.serrulata var.serrulata is one species of indigenous tree distributed widely in China which has a large potential of wide application because of its high value of ornamental,strong adaptability and strong resistance.C.cerasoides var.rubea is one variation of C.cerasoides,it’s a rare species flowering in winter which flower is semidouble and rosy red,but used less in landscape architecture,will have a great market prospect.In this study,the stem segments of C.yedoensis ‘Somei-yoshino’,C.serrulata var.serrulata and C.cerasoides var.rubea,mature embryos of C.yedoensis ‘Somei-yoshino’ and C.serrulata var.serrulata were used as explants.Screening induction medium,multiplication medium and rooting medium,and the transplanting technology of test tube seedlings were studied in order to construct a perennial technique of seedings production.The main findings as follows:1.The best fit sterilization method for unlignified stem with axillary bud and embryo is 75% alcohol for 30 s and 2% Na Cl O for 300s;The best fit sterilization method for stem with axillary bud of half lignification is 75% alcohol for 30 s and 0.1% Hg Cl for 420 s.2.In early spring,the best explant is unlignified stem with axillary bud,stem with axillary bud of half lignification has high contamination rate and high browning rate,the frequency of induction was decreased.After the cherry matures,the best explant is embryo,the contamination rate is extremely low and to produce clustered shoots.3.For C.yedoensis ‘Somei-yoshino’,MS medium supplemented with 2.0mg/L 6-BA and 0.1mg/L IBA showed the highest single bud induction rate(61.29%)using stem with axillary bud as explants,MS medium supplemented with 2.0mg/L6-BA,0.05mg/L IBA and 0.05mg/L NAA showed the highest single bud induction rate(86.02%)and to produce clustered shoots by using embryo as explants.For C.serrulata var.serrulata.MS medium supplemented with 1.5mg/L 6-BA and 0.2mg/L NAA showed the highest single bud induction rate(87.10%)using stem with axillary bud as explants,MS medium supplemented with 2.0mg/L6-BA and 0.1mg/L IBA showed the highest single bud induction rate(86.02%)and to produce clustered shoots by using embryo as explants.For C.cerasoides var.rubea,MS medium supplemented with 1.5 mg/L 6-BA and 0.1 mg / L IBA showed the highest single bud induction rate(47.05%).4.For C.yedoensis ‘Somei-yoshino’,MS medium with 2.0mg/L6-BA and 0.1mg/L IBA exhibited the highest clustered bud proliferation rate(3.73 and 4.09).For C.serrulata var.serrulata,MS medium with 2.0mg/L6-BA,0.05mg/L IBA and 0.05mg/L NAA exhibited the highest clustered bud proliferation rate(5.37 and 5.67).The results show the difference of explant was not a significant factor to clustered bud proliferation rate.For C.cerasoides var.rubea,MS medium with 2.0 mg / L 6-BA,0.15 mg / L IBA and 0.15 mg / L NAA exhibited the highest clustered bud proliferation rate(1.34).5.For C.yedoensis ‘Somei-yoshino’,Half MS medium with 0.5mg/L IBA and 0.1mg/L NAA was suitable for plantlet rooting,and the rooting rate was 70.97%.For C.serrulata var.serrulata,Half MS medium with 0.8mg/L IBA and 0.1mg/L NAA was suitable for plantlet rooting,and the rooting rate was 96.77%.For C.cerasoides var.rubea,Half MS medium with 0.6 mg / L IBA was suitable for plantlet rooting,and the rooting rate was 81.07%.6.When the plantlets were due to be moved to the nursery.The medium with 50% humus,25% sand and 25% roseite was effective for rooting.Three kinds of subjects all showed the highest survive rate.For C.yedoensis‘Somei-yoshino’,the survive rate was above 87.09%.For C.serrulata var.serrulata,the survive rate was above 77.42%.For C.cerasoides var.rubea,The survive rate was above 33.33%. |
