Macleaya cordata(willd)R.Br and Macleaya microcarpa(Maxim.)Fedde are belong to Papaveraceae family and more than 100 isoquinoline alkaloids were reported from Macleaya species.The chemical constituents of M.cordata and M.microcarpa are highly similar,but at present,the most researches of Macleaya species mainly focuses on the isoquinoline alkaloids of M.cordata,while there a few studies on M.microcarpa and other chemical compounds.In this study,we mainly studied the compounds in fresh stem and leaves of M.microcarpa in early flowering stage by HPLC-Q/TOF-MS.According to the analysis results,preferential separated the components containing more unknown components by silica gel column chromatography,recrystallization and high performance liquid chromatography et al.We got 13 compounds from the M.microcarpa.The structures of the 13 compounds were identified by NMR(1H-NMR,13C-NMR),2D-NMR(1H-1H COSY,HMBC,HSQC)and mass spectrum(HRMS,HRMS/MS).The result as follows: alkaloids: dihydrosanguinarine(1),ethyl 2'-(5,6-dihydrosanguinarine-6-yl)acetate(2),ethyl 2'-(5,6-dihydrosanguinarine-6-yl)formate(4),ethyl 2'-(5,6-dihydrocheletrythrine-6-yl)acetate(5),sanguinarine(9),6-(2-hydroxyethyl)dihydrochelerythrine(10),methyl(5,6-dihydrosanguinarine-6-yl)acetate(11),ethyl(5,6-dihydrosanguinarine-6-yl)acetate(12),iso-ethyl(5,6-dihydrosanguinarine-6-yl)acetate(13).Phenylpropanoids: 3-O-feruloylquinic acid(6),4-O-feruloylquinic acid(7),5-O-feruloylquinic acid(8),and steroids: stigmasterol(3).Compounds 11,12 and 13 are less than 1 mg,so they are inferred by mass spectrometry pyrolysis.Compound 4 is new compound.Compound 11,12 and 13 are inferred as new compounds.Compound 2,5,7 and 8 are isolated from the genus of Macleaya for the first time.Compound 10 are isolated from this plant for the first time.
