In dairy farming,cow mastitis caused by pathogenic infections,improper management,environmental abnormalities,genetic factors and other factors is the most common disease in dairy farming,which also caused the most economic losses.Cow mastitis leads to reduced milk production and poor milk quality,which can severely cause breast necrosis and even endanger the life of the cow.Currently,antibiotics are often used clinically to treat cow mastitis,but there will be adverse consequences of antibiotic residues and increased microbial resistance.This study explored a new treatment method for cow mastitis,in order to treat mastitis without side effects.In this study,combined with the pathogenesis of cow mastitis,the author designed a compound Chinese herbal medicine additive and fed it to the cow to observe the effect of Chinese herbal medicine on cow mastitis,in order to treat cow mastitis.This experiment is divided into an in vivo test and an in vitro test.In vitro test:in this study,bovine mammary epithelial cells were used as in vivo model to treat cow mammary epithelial cells with different concentrations of effective components of Chinese herbal medicine.In vivo test:in this study,20 normal cows and 20 recessive mastitis cows(the number of somatic cells in milk is more than 5×10~5/ml)were selected respectively,and the normal cows were randomly divided into two groups,the blank control group and the prevention group,each group was 10,the blank control group was fed with basic diet,and the pre prevention group was fed with basic diet with compound additives;Mastitis cows were randomly divided into two groups,the treatment group and the negative control group,10 cows in each group.The whole feeding experiment lasted for 30 days.Milk samples were collected every 7 days to detect the number of somatic cells,the rate of milk fat and the rate of milk protein.Blood was collected from tail vein every 15 days to detect the changes of SAA,IL-1?,IL-2 and IL-8 in serum.Result:(1)Test 1:Different concentrations of active components of Chinese herbal medicine have no toxic effect on the cell viability of bovine mammary epithelial cells.The cell viability of bovine mammary epithelial cells can be significantly improved when the concentration of chlorogenic acid(dandelion anti-inflammatory active substance)is 10?g/ml,the concentration of sodium Houttuyniae element is 1?g/ml,and the concentration of flavonoid glycoside of Herba Vaccariae is 20?g/ml.(2)Test 2:There was no significant change in the number of somatic cells between the control group and the prevention group.From the 14th day of feeding,the difference between the treatment group and the blank control group decreased gradually.On the 21st day,the difference between the treatment group and the normal group was not significant(P>0.05).In contrast,the number of somatic cells in the negative control group without Chinese herbal medicine was kept at a high level,and significantly higher than that in the normal group(P?0.05).(3)Test 3:The results showed that throughout the feeding experiment,there was no significant change in the milk protein rate and milk fat rate of cow milk in different groups and different treatments(P>0.05).(4)Test 4:The SAA of the treatment group was significantly different from that of the first day(P<0.05),and returned to the normal level;the SAA of the blank control group and the prevention group had no significant change(P>0.05).There was no significant change of IL-1?in the serum of the control group and the prevention group(P>0.05).The IL-1?in the treatment group on the 30th day was significantly lower than that in the early stage of treatment(P<0.01).On the 15th and 30th day,the level of IL-2 in the treatment group was significantly lower than that in the initial treatment group(P<0.01),and the level of IL-2 in the prevention group was significantly lower than that in the blank control group(P<0.05).After 30 days of experiment,the level of IL-8 in the serum of the treatment group was significantly lower than that of the first day(P<0.05),and Chinese herbal medicine had no significant effect on the level of IL-8 in the prevention group(P>0.05).Conclusion:Compound Chinese herbal medicine additives have certain preventive and therapeutic effects on cow's recessive mastitis.
