The recalcitrant loquat seeds are sensitive to low temperature and dehydration,they can not be dried and stored in low temperature for a long time.however,for sowing and seedling,it generally necessary to pick and sow seeds in the same year.At present,physiological and biochemical variation laws under different storage conditions after seeds harvestd,as well as the storage and transportation conditions and time limits for keep viability which are all not clear.In order to explore the internal causes of physiological and biochemical index change and loss of seed viability after seeds were picked,to provide therotical basis for effective application in production in greatest extended.In this research,the'Changqizaosheng'and'Changshengzaosheng'loquat seeds were used as study objects,to study the changes of physiology and biochemistry during seeds storage at room temperature,17?and 10?.Loquant seed vigor when stored at17?for 12d was higher,and then showed down trend;the germination rate was above 60%at 35d,which was significantly higher than at 25?and 10?.Seeds water content declined with the increase of storage temperature,'Changqizaosheng'water content of each temperature stored at 35d was very significant,'Changshengzaosheng'water content of room temperature stored at 35d was significantly different from 17?and 10??P<0.01?.With the increase of storage time the relative conductivity of two species increased,and the membrane oxidation product MDA gradually accumulated.With the increase of storage time,the germination rate and water content of two loquant varieties at three storage temperatures gradually declined,the relative conductivity and MDA content gradually increased,the activities of protection enzymes SOD and POD showed a slowly downward trend throughout the process,the basic nutrients of total solube sugar,reducing sugar,starch content increased first and then declined.The contents of ZR and ABA during the radicle elongation of the two seeds increased first and then gradually decreased;the content of IAA gradually increased.Correlation analysis showed that the germination of'Changqizaosheng'seeds was significantly and positively correlated with water content and cell protection enzyme SOD activity,and was significantly and negatively correlated with relative conductivity and MDA content.The indicators that have a significant positive correlation with germination rate of'Changshengzaosheng'were water content and soluble sugar,and the crude protein content is significantly negatively correlated.The analysis of absolute and relative content of hormones showed that the ratio of IAA/ABA??GA3+IAA?/ABA in radicle of'Changqizaosheng'gradually increase when it was 0?7mm long,and then begin to decline.the peak value of IAA/ABA??GA3+IAA?/ABA of'Changshengzaosheng'came in 3mm and 10mm of radicle;in two varieties the content of IAA and ABA significantly higher than GA3 and ZR.In conclusion,the storage effect of both varieties at 17?is better than room temperature and 10?,the process of seed germination and radicle enlongation was mainly caused by interaction of hormones IAA,ZR and ABA.When preservation seeds,consider reducing the degree of cell membrane oxidation,maintaining intergrity of the cell structure,and reducing cell respiration,try to use the seeds before they reach critical water content.It is recommended to choose seeds of good quality to be stored at about 17?for about one month.Seeds stored as germplasm can be improved by adding exogenous IAA and ZR during germination.Seeds stored as raw materials for processing can be stored for a short period of time until the content of avaliable nutrients is higher.
