Study On The Immune Activity And Antitumor Activity Of Shengmai-san Ethanol Extract

As one of the famous ancient prescriptions of traditional Chinese medicine in China,Shengmai-san is also known as Shengmai-yin,which originated in Jin Dynasty Zhang Yuansu's medical book "medicine origin".Shengmai-san mainly used in the clinical treatment of coronary heart disease,heart failure,acute myocardial infarction and other cardiac and cerebral vascular diseases.However,the immune and anti-tumor activity are still unknown.Here,we use anhydrous ethanol as solvent,adopt the method of reflux extraction to prepare Shengmai-san ethanol extractct,confirm the total saponin as a symbol of the composition of Shengmai-san ethanol extractct and determine the content.Preliminarily evaluate the toxicity of the extracts,in accordance with the method of enhancing the immunity function in the technical standards for testing and assessment of the health food in version 2003,we use the female Kunming mice as experimental objects.The amount is 1,2,6g/kg three dosage groups of Shengmai-san ethanol extractct,control group(distilled water)and thymosin group(10mg/kg)were given by intragastric administration respectively,once a day for 30 days.Determination of the viscera index,cellular immune function,humoral immune function,macrophage monocyte function and NK cell activity respectively.Researching the immune activity of Shengmai-san ethanol extractct for the system.Determined by MTT colorimetric method is used to evaluate Shengmai-san ethanol extractct are activated to restrain activity of NSCLC cell excessive activation of the ras gene.The conclusions are as follows:1.Using anhydrous ethanol as solvent reflux extraction to prepare Shengmai-san ethanol extractct.For the first time with 10 times the size of anhydrous ethanol,and the second time with 8 times the size of anhydrous ethanol,each time lasts 2 hours.2.Determine the total saponin as signature ingredient of the Shengmai-san ethanol extractct.By UV visible spectro-photometric method to measure the content of total saponin in Shengmai-san ethanol extractct is 2.69g/100g.3.Acute toxicity experiment shows that the maximum dissolved concentration of Shengmai-san ethanol extractct is 371 mg/ml,to the Kunming mice by maximal tolerance dose of gastric Shengmai-san ethanol extractct is 7.42g/kg.4.Comparing with the control group,Shengmai-san ethanol extractct could significantly stimulate the proliferation of mice spleen lymphocyte transformation function,promote the mice delayed hypersensitivity reaction and enhance monocyte macrophages of the mice carbon clearance rate.However,there is no effect on the rest of the detection index.The results reveal Shengmai-san ethanol extractct could enhance immune function.5.Application of MTT colorimetric assay of Shengmai-san ethanol extractct on the inhibition of lung cancer cell line which ras gene mutations,finding that the IC50 of ras NSCLC cells H1299,A549 and H358 cell in 48h is 82?g/ml,130?g/ml and 249?g/ml respectively.Shengmai-san ethanol extractct could inhibit cell proliferation in a dose-dependent and time-dependent manner in vitro,and the sensitivity is H1299>A549>H358.Revealing that Shengmai-san ethanol extractct has an inhibition ability on NSCLC cells,but has no selectivity on ras NSCLC cells.
