FIVE1 which is emerged in recent years is suitable for the whole animal medical researches and vivo histological study as a tool.Intravenous injection or spraying fluorescence imaging agents on its surface can realize the functions and molecular imaging,trends and adjustable imaging depth control.It is a kind of real-time trends to observe the tissue,and reduces the artificial factors when the traditional producing biopsy that pathological observation can be brought into.The traditional diagnostic method of RA relies on destroying the standard pathological histology technology to provide a basis.This needs to put animals model to death,and bring into the artificial factors in the process of making the pathological biopsy.The introduction of FIVE 1 have made it possible to observe the living bodies.We can observe articular cartilage form in current time through intravenous injection or spraying fluorescent contrast agents on the mucous membrane surface.It can break a new path for the diagnosis and treatment of rheumatoid arthritis.Objective:This topic puts the rabbits as models,and evaluated the rabbits'articular cartilage histological between the normal rabbits and the rheumatoid arthritis ones based on FIVE 1 and optical microscope.We selected the adjuvant induced arthritis and ovalbumin rheumatoid arthritis as models.And we use the same animal model to carry on the traditional pathological diagnosis.It made a further verification for the authenticity and reliability through the FIVE1 for related tissue.Due to the related literature at home and abroad is rare,our exploration of rabbit articular cartilage histological based on FIVE1 is a preliminary exploration and provides the researchers later with the preliminary methodology for RA diagnosis and treatment.Methods:This research firstly lighten on the knee joints and plantar tissue of the normal rabbits with the fluorescence isothiocyanate.And study and evaluate the several of tissues and organs with FIVE1,including cartilage cells,synovial tissue,meniscus,tendons,connective tissue,muscle tissue and so on.Then,diagnose the knee joints and plantar joints of the normal rabbits with the conventional pathological diagnosis.The process of the making the pathological slices includes decalcification,dehydration,lucency,paraffin,embedding and dye.Secondly,we prepare for the animal models with RA.The selected models are rabbits with adjuvant induced arthritis and ovalbumin arthritis.After molding,we successfully created the rheumatoid arthritis models what we demand of through the observation on the index with the cross-section diameter of ankle wrist,the diameter of the knee joint,the change of weight,and change of food-intake index.We applied FIVE I to carry on the histological assessment on the metatarsus of adjuvant arthritis rabbit and the knee articular cartilage of the ovalbumin arthritis rabbit.Then compare them with the normal rabbits.Finally,we make pathological slices on the metatarsus and the knee articular cartilage of the animal models with adjuvant arthritis and ovalbumin arthritis above.And we began to evaluate them by optical microscope.Results:There have been many reports about the adjuvant induced arthritis with the rats as models.However,there are less using rabbits as models.This topic selected rabbits as the experimental object to make the adjuvant induced arthritis.It has been proved that we have successfully caused the adjuvant induced arthritis through the measurements of the cross-section diameter of rabbits' knee,ankle and wrist,active state,food-intake index.In this study,we observed with several index such as the ovalbumin before and after sensitization,the consumption of feed quantity,the diameter of knee joint and the change of weight index.The success rate of ovalbumin arthritis model is about 70%.Under the FIVE1,the histological features have obvious distinction on the knee joint cartilage cells between the ovalbumin arthritis rabbits and normal rabbits.It is the comparison of defective of chondrocytes and normal ones.The patholog,ical changes of articular cartilage have damaged,erosion,and the cell borders have blurred.Part of the chondrocytes is missing which is in accord with the pathological diagnosis.Conclusion:l.The preparation of adjuvant induced arthritis and ovalbumin arthritis models are successful.2.Imaging system assessed the features of histological morphology on the articular cartilage objectively,real-time and effectively.In this topic,the application of FIVE1 made an accurate assessment on the histological characteristics with the knee joint cartilage and metatarsus cartilage of the model rabbits and the normal rabbits.3.We successfully to have a pathological diagnosis on the knee joint cartilage and metatarsus cartilage of the model rabbits and normal rabbits with the help of optical microscope.
