Objective:To investigate the effect and mechanism of GuanXin SuHe capasules against acute myocardial ischemia injury.Methods:① Establishment the model of acute myocardial infarction in rats.The model rats were randomly divided into five groups and respectively treated with pure water,high-dose GuanXinSuHe(0.8g/kg),low-dose GuanXinSuHe(0.4g/kg),captopril for 5 days.Determined cTn-T,AST,CK,CK-MB after ligating 48h.②Preparation of the serum containing GuanXinSuHe(0.8g/kg)with Serum Pharmacological Method.Cardiocytes derived from neonatal rats that cultured in Dulbecco Modified Eagle Medium(DMEM).The cardiocytes were injured with hydrogen peroxide,and then treated with the serum containing different concerntrations of GuanXinSuHe.The survival rate of cardiocytes was measured with MTT method,and determined MDA,LDH,T-SOD,CAT of myocardial cells.③Specimens of isolated thoracic aorta of the rat were prepared and subjected to the following experiments:Direct effect of GuanXinSuHe capsule on the aorta;Effect of GuanXinsuhe capsule on the contraction of the aorta induced by norep inephrine(NE)or potassium chloride(KC1).Results:①The area of myocardial infarction in the GuanXinSuHe high dosage group was reduced,cTn-T and AST decreased(P<0.05,P<0.01,P<0.01)-The effect of 0.8g/kg GuanXinSuHe and 0.4g/kg GuanXinSuHe on ECG in acute myocardial infarction rats was observed,the heights of ST segment were reduced just after ligating Oh and 48h.②The neonatal cardiocytes were injured with H2O2 for 2h,and the survival rates were decreased significantly compared with normal cells(P<0.01).The content of MDA and LDH were increased significantly,but the activity of SOD and CAT were decreased.After treated with the different concentrates of drug-containing serum,the survival rate was increased with a dose dependant manner.The content of MDA and LDH were decreased and the activity of SOD and CAT were significantly increased(P<0.01).③ GuanXinSuHe capsule in various concentrations had direct contraction effect on the aorta,especially in 100-200mg/L,GuanXinSuHe capsule compared with blank control group had significant difference(P<0.05);GuanXinSuHe capsule in 200mg/L significantly inhibited the contraction of the aorta induced by NE and KCL(P<0.05).Conclusion:①GuanXinSuHe has significant effect on myocardial infarction,It treats myocardial infarction through regulating cTn-T and AST.②GuanXinSuHe capsules can protect the cardiocytes from H2O2 injured by increasing antioxidation capability.③ GuanXinSuHe capsules was shown that,it not only directly contract the isolated rat thoracic aorta,but also inhibited the contraction of theaorta induced by norep inephrine and potassium chloride.
