Objective To determine the expressions of matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP-1)in the tissues of intestinal adhesion,and explain the preventing intestinal adhesion mechanism about ligustrazine.Methods Seventy four SD male rats were randomly assigned to four groups:normal group(n=18).sham operation group(n = 18),operation group(n = 18)and drug group(n = 18).In the operation and drug group,the serous membrane of ileum was injured via scrapping;the dehydrated alcohol was dropped on the surface of wound;cecal mesenteric artery was clamped;so themodel of traumatic intestinal adhesion of rat was established.In the sham operation group,the anesthesia and laparotomy were same as operation group,but no intestinal tissue was injured,and no mesenteric artery was clamped.In drug group,every day rats were intraperitonealiy injected ligustrazine,while in the rest groups equivalent physiological saline were injected,which were not completed until they were sacrificed.In the four groups,6 rats were respectively sacrificed at 2 d,7 d,14 d after modeling,and the tissues of serous membrane in adhesive area were taken.Under macroscopic observation,intestinal adhesion were classified.The expressions of MMP-9 and TIMP-1 in the intestinal adhesive tissues and the normal intestinal tissues were determined by immunohistochemistry assay and image analysis.Results(1)The statistics of intestinal adhesion degree classification:Compared with the surgical,comparing with sham operation and normal group,the intestinal adhesion degree in operation and drug group increased significantly(P<0.05);Compared with the operation group,the drug group was significantly reduced(P<0.05).(2)Compared with control group and 2 days after modeling in observation group,the MMP-9 expression reduced,but the TIMP-1 expression elevated to the peak value at 7 days after modeling in observation group(all P<0.01).Compared with operation group and 2 days after modeling in drug group,the expressions of MMP-9 dropped(P<0.05),moreover,the TIMP-1 were no significant difference.(P>0.05)In 7 or 14 days after modeling in drug group,the expressions of TIMP-1 increased(P<0.05),however,the MMP-9 were not obviously different.(P>0.05).Conclusions Through the cell factor including the expressions of MMP-9 and TIMP-1 in the intestinal tissues,to a certain extent,ligustrazine can inhibit the rat intestinal adhesion.
