Objective: Topreliminarilystudy the expression of enolase-1(ENO-1)in saliva and tissuesboth in normal people and patients with oral squamous cell carcinoma(OSCC),and further explore the correlation between the expression of enolase-1 and oral OSCC.Methods:(1)In order to find the difference of ENO-1 expression between normal tissues and OSCC tissues.(2)The expression of ENO-1 in saliva both in normal population and OSCC was detected by Enzyme-linked immunosorbent assay(Elisa).(3)The protein imprint test(Western Blotting,WB)was used to detect the difference in the expression of ENO-1 in normal tissue and OSCC tumor tissue.(4)The results of IHC and ELISA were analyzed by T test and the outcome of Western Blot were analyzed by rank sum test.Additionally,the relationship between the change of expression of ENO-1 both in saliva and tissues was examined by Pearson correlation test.Results:(1)The outcome of IHC showed that ENO-1 is expressed in both normal tissues and OSCC tissues.However,the expression of ENO-1 in OSCC was significantly higher than that in normal group,and the difference was significant(P< 0.05).In normal tissues,the expression of ENO-1 was weakly positive and a pale yellow or brown yellow granule in the cytoplasm can be observed under the microscope.Compare the normal tissues,ENO-1 was positive or strongly positive expression was in OSCC tissues,and we can see deeper yellow or pale brown granules in the cytoplasm with microscope.Furthermore,there was no significant difference in the expression of ENO-1 in the OSCC tissues of different pathological grades(P >0.05).(2)The expression of ENO-1 in saliva of OSCC patients was significantly higher than that of normal people,and the statistics difference was significant(P<0.05).(3)What's more,the results of WB illustrated that: there was a significant difference in the expression of ENO-1in OSCC tissues and normal tissue,put it another way,the expression of ENO-1 in OSCC was much higher than that in normal tissue and the statistical difference was significant(P>0.05),whichsuggests that the changes of the expression of ENO-1 may be associated with the occurrence of tumor and the increase of ENO-1 expression may raise the risk of OSCC.(4)Finally,Pearson correlation test was used to analyze the relevance of the expression of EN0-1 between in saliva and tumor tissues,and the results display that ?=0.37,P=0.174,which indicated that the changes of ENO-1 in the tumour tissue are related to the changes of saliva.The results suggest that the changes of ENO-1 in the saliva can replace the detection of ENO-1 in the tissues to monitor the occurrence of OSCC.Conclusion:(1)The expression of ENO-1 in the tumour tissues and saliva of the patients with oral squamous cell carcinoma was highly expressed,and the changes of the two were consistent.(2)Detecting the expression of ENO-1 in saliva is simple and convenient,which is helpful for early screening and early diagnosis of oral squamous cell carcinoma.
