Object: To investigate the expression of N-?-acetyltransferase 10 protein(Naa10p),Carcinoembryonic antigen(CEA)of saliva and serum in Oral Squamous Cell Carcinoma(OSCC)patients,precancerous lesions and normal adults,and to explore the relationship between the development of Naa10 p,CEA and oral squamous cell carcinoma and the diagnostic value of combined detection in oral squamous cell carcinoma.Methods: The expression of Naa10 p,CEA in saliva and Naa10 p in serum of 76 patients with oral squamous cell carcinoma,30 patients with precancerous lesions and 40 normal subjects were detected by ELISA.And to analyze the correlation between the expression of Naa10 p in saliva and serum and the clinical parameters such as age,sex,ethnicity,differentiation degree,clinical stage and tumor metastasis in oral squamous cell carcinoma.The Receiver Operating Characteristic(ROC)curve was drawn with the results of the pathology test to evaluate the diagnostic performance of Naa10 p and CEA of saliva in the diagnosis of oral squamous cell carcinoma.Results:(1)The expression of Naa10 p and CEA in saliva of oral squamous cell carcinoma group,precancerous lesion group and normal control group were statistically significant(P < 0.05).The expression of Naa10 p in oral squamous cell carcinoma was significantly higher than that in normal control group,whether serum or saliva.The expression of Naa10 p in saliva was significantly higher than that in serum,both in oral squamous cell carcinoma and normal control group.(2)The expression of Naa10 p in saliva of oral squamous cell carcinoma was correlated with the degree of differentiation,and the expression of Naa10 p in serum was related to age,the expression of CEA in saliva was correlated with clinical stage and lymph node metastasis(P <0.05),and the others were not statistically significant(P>0.05).(3)The area under the ROC curve of Naa10 p in saliva(0.924)was significantly higher than that in serum(0.713),and the difference was statistically significant(P < 0.05).The sensitivity,specificity and accuracy of Naa10 p in saliva and Naa10 p in serum were(85.40%,85.00%,85.30%),(74.20%,65.00%,72.10%).The sensitivity,specificity,accuracy,positive predictive value and negative predictive value of Naa10 p,CEA single detection and combined detection were(81.70%,72.50%,78.00%,81.70%,72.50%),(88.30%,47.50%,72.00%,87.60%,86.90%)and(91.70%,80.00%,87.00%,87.30%,86.50%);The area under the ROC curve of Naa10 p,CEA and combined detection in saliva was0.822,0.736 and 0.911,respectively.The area under the ROC curve of the combined detection was greater than that of any single test(P < 0.05).Conclusion:(1)Naa10p is highly expressed in saliva and serum of oral squamous cell carcinoma.The expression of Naa10 p in saliva is related to the degree of tumor differentiation,suggesting that Naa10 p may be related to the development of oral squamous cell carcinoma,And the expression of Naa10 p in saliva was higher than that in serum,suggesting that the detection of Naa10p in saliva was more clinically significant than the detection of serum for oral squamous cell carcinoma.(2)CEA in oral squamous cell carcinoma saliva in the high expression,indicating that saliva CEA can also help the diagnosis of oral squamous cell carcinoma.(3)The combination of Naa10 p and CEA in saliva helps to improve the diagnostic performance of oral squamous cell carcinoma,which is an ideal biomarker for oral squamous cell carcinoma.
