Objective: Observe the effects of Zhuang-medicine medicated-thread moxibustion on blood glucose,urine glucose,the gastric emptying rate,small intestine propulsive rate,Gastric growth promoting hormone(Ghrelin),cholecystokinin(CCK),stem cell factor(SCF)and the expression of c-kit positive cells(ICCs)in diabetic gastroparesis(DGP)model rats.Taking the signal pathway of SCF/c-kit and brain intestinal peptide Ghrelin?CCK as the point of penetration to discusse the therapeutic mechanism of moxibustion with the ignited Zhuang-medicine for regulating blood glucose,improving gastric emptying and treating DGP.Methods: 80 SD rats were randomly divided into 4 groups: normal group,DGP model group,Zhuang-medicine medicated-thread moxibustion group(medicated-thread moxibustion group),and metoclopramide group,20 rats in each group.DGP model was established with intraperitoneal injection Streptozotocin(STZ)and in combination with a high-sucrose/high-fat diet for 8 weeks.The moxibustion group was treated with moxibustion of the moxibustion group,and the gastric complex group was treated with 1.7% gastro-compound ampoule(1mL/100g),which was all 1 times/d,with a continuous 15 d.The medicated-thread moxibustion group was treated by Zhuang-medicine medicated-thread moxibustion,and 1.7% metoclopramide(1mL/100g)was given to rats of the medication group,once daily,for 15 days.After treatment,blood glucose,gastric emptying rate and small intestinal propulsive rate were measured.The Ghrelin and CCK in gastric antrum were determined by ELISA.The expression of c-kit positive cells(ICCs)in the gastric antrum was detect ed by Immunohistochemical method.Results:(1)Diet volume and body weight: compared with the nor mal group,the body weight and diet volume of the model group decre ased significantly(P<0.01);compared with the model group,the body weight and diet of the medicated-thread moxibustion group increased si gnificantly(P<0.01).(2)blood glucose and urine glucose: compared with the normal group,the blood glucose and urine glucose of the model gr oup increased significantly(P<0.01),indicating that the DM model was es tablished successfully;compared with the model group,the blood gluco se and urine glucose of the medicated-thread moxibustion group decrea sed significantly(P<0.01).(3)Gastrointestinal propulsive rate: compared w ith the normal group,the gastric emptying rate and the intestinal propul sion rate of the model group were significantly decreased(P<0.01),whi ch indicated that the gastrointestinal propulsion rate of the DM model r ats decreased,combined with the symptoms and blood glucose values,i ndicating the model of DGP rats was successfully established;and com pared with the model group,the gastric emptying rate and intestinal pr opulsion rate in the medicated-thread moxibustion group were significa ntly higher than those in the metoclopramide group(P<0.01).(4)The lev el of Ghrelin and CCK: compared with the normal group,the level of Ghrelin decreased and the content of CCK increased in the model grou p(P<0.01);compared with the model group,the level of Ghrelin increa sed and the content of CCK decreased in in the medicated-thread moxibustion group(P<0.01).(5)Content of SCF and the expression of c-kit positive cells: Compared with the normal group,the model group were significantly decreased(P<0.05),whereas those indexes of the model g roup were significantly lower than those in the medicated-thread moxibu stion group(P<0.05).The content of SCF and the expression of c-kit p ositive cells were increased in the medicated-thread moxibustion grou p(P<0.05).Under optical microscope,the expression of c-kit positive p rotein in normal group was more than that in metoclopramide group,a nd the c-kit positive protein of model group was significantly decreased and scattered in muscle layer.Conclusion: The zhuang-medicine medicated-thread moxibustion can regulate gastrointestinal peristalsis and function,and increase the di et and body mass of DGP rats.The zhuang-medicine medicated-thread m oxibustion regulates the blood glucose of DGP rats by directly stimulati ng pancreatic beta cells,promoting insulin secretion,and lowering blood sugar level.The mechanism of Conclusion Zhuang-medicine medicatedthread moxibustion on improving gastrointestinal motility in DGP rats is through(1)raised serum Ghrelin level and lower gastric antrum CCK le vel,correct the disorder state of brain gut peptide in peripheral andbrai n(2)Upregulation of SCF/c-kit signaling pathway,reversing the apoptosis and degeneration of ICCs and restoring the gastric electric rhythm of SMC;These may be the mechanism of the treatment of DGP with the moxibustion with ignited Zhuang-medicine medicated-thread therapy...
