Experimental Observation Of Xiao Tan Di Yin Tongzhi Formulation Affecting Rat Model Relevant To Schizophienia With Tan Shi Symptom

Objective:This experiment was designed to explore a method of constructing one kind of "combination of disease and symptom" model rats relevant to Schizophrenia with tan shi symptom,to observe "combination of disease and symptom" model rats about their behavioral indicators,body weights,body lipid content indicators,serum total cholesterol and triglyceride levels.At the same time,the formulation of Xiao Tan Di Yin Tong Zhi was used to affect the "combination of disease and symptom" model rats and the results were recorded in this paper.Methods:In this research,adult male SD rats were randomly divided into blank group,model group and intervention group.Rats of model group and intervention group were fed with high-fat diet for 2 weeks to gain weiht and to obtain high blood lipid,at the same time,drug MK-801,as a tool drug for establishing rat model of schizophrenia,was used to build the model with intraperitoneal injection for 2 weeks in model group and intervention group.During the establishment of the model,intervention group was gavaged with Xiao Tan Di Yin Tong Zhi formulation and model group was gavaged with distilled water of the same capacity weight ratio.During the establishment of the model,blank group rats were fed with common feed,intraperitoneal injected with 0.9% sodium chloride solution with the same volume weight ratio,and gavaged with distilled water of the same capacity weight ratio.The effects of learning and memory ability and spatial recognition on cognitive abilities of Morris water maze in each group of rats were observed.The effects of exploratory activities and stereotype behaviors in each group of test rats were observed,and the results in each group were measured including body weights,body lipid content,serum total cholesterol and triglyceride levels.Results:1.In water maze experiment,compared with the blank group,the escape latency of the model group increased,and the difference was statistically significant(P<0.05).The value number of crossing the original platform area decreased,and the difference was statistically significant(P<0.01).The distance ratio of central activity decreased,and the difference was statistically significant(P<0.01).The time ratio of central activity decreased,and the difference was statistically significant(P<0.05).Compared with the model group,the escape latency significant(P<0.01).The grooming times of the model group incresased,and the difference was statistically significant(P<0.05).Stools of the model group incresased,and the difference was statistically significant(P<0.05).Compared with the model group,the total distance value number in intervention group increased,and the difference was statistically significant(P<0.05).The grooming times of intervention group decreased,and the difference was statistically significant(P<0.05).Stools of of intervention group decreased,and the difference was statistically significant(P<0.05).3.Compared with the blank group,the value of body weight of model group was significantly higher and the difference was statistically significant(P<0.01);The value number of body lipid content around testicles and kidneys increased,and the difference was statistically significant(P<0.01).Compared with the model group,body weight in intervention group were decreased,and the differences was statistically significant(P<0.05);Body lipid content around testicles and kidneys in intervention group decreased,and the differences was statistically significant(P<0.01).4.Compared with the blank group,the value number of serum total cholesterol(TC)was higher in the model group,and the difference was statistically significant(P<0.01).The value numbers of triglyceride(TG)was higher in the model group and the difference was statistically significant(P<0.01).Compared with the model group,the value number of TC was lower in intervention group and the difference was statistically significant(P<0.01);the value number of TG was lower in intervention group and the difference was statistically significant(P<0.01).Conclusion:1.The "combination of disease and symptom" model rats had less exploratory activities,more stereotyped movements and tension movements,poorer spatial recognition ability and memory ability.And it could show the behavioral symptoms of schizophrenia.The established "combination of disease and symptom" model rats gained body weight and body lipid content,and the measured serum lipids were increased.In line with the rules of modern medical laboratory indicators,this experiment could reach the purpose of modeling in TCM.2.The formulation of Xiao Tan Di Yin Tong Zhi can improve the behavioral symptoms of rat model relevant to "combination of disease and symptom",can improve spatial recognition ability and memory ability,reduced stereotyped movements and tension movements,can reduce the body weight and body lipid content around testicles and kidneys,and can reduce the serum lipids levels.It could show the effect of Xiao Tan Di Yin Tong Zhi formulation in treatment relevant to this “combination of disease and symptom” model.
