Objective To provide evidence for diagnosis,staging,evaluation of prognosis and gene therapy of prostate cancer(PCa), the expression of hypoxia-inducible factor 1α(HIF-lα),vascular endothelial growth factor(VEGF) and glucose transporter-1 (GLUT-1) mRNA in flash tissues of prostate cancer and benign prostate hyperplasia (BPH) was detected quantitatively, and the relationship and clinical significance of them was investigated conneting with the clinicopathological characteristics.Methods The expression of HIF-1α,VEGF and GLUT-1 mRNA in PCa(18 cases) and BPH(24 cases) were detected by real-time fluorsescent quantitative reverse transcription-polymerase chain reaction (FQ-PCR).Among the 18 cases,there were 8 cases in clinical stageⅡ,3 cases in stageⅢ, 7 cases in stageⅣand 1 cases in histological grade 1,6 cases in grade 2,11 cases in grade 3-4.We study the relationship between the expression of them and the clinicopathological characteristics,and we also assessed the mutual correlation of the three markers in PCa.Results In the 18 specimens of PCa ,HIF-1αmRNA was expressed in 12 of them. The expression levels in PCa(median:2.4894) were higher than those in BPH..The differences between them were statistically significant(P=0.0005). The same to GLUT-1 mRNA,and the median of the expression levels in PCa was 4.8006. VEGF mRNA was expressed in 13 of them. The expression levels in PCa(median:5.5162) were higher than those in BPH.The differences were also statistically significant(P=0.0118).The higher clinical stages and histological grades the PCa was ,the higher the expression levels of HIF-1α,VEGF,GLUT-1 mRNA was. The mean expression levels in stagesⅢ+Ⅳwere higher than those in stagesⅡ,the same to grades G3-4 and G1+G2, and the differences among the different pathological grades and clinical stages were statistically significant(P <0.05). The expression of HIF-1αmRNA was positive relation with the VEGF and GLUT-1 mRNA(r1=0.5916,P=0.0097;r2=0.6510,P=0.0034). The expression of VEGF mRNA was also positive relation with the GLUT-1mRNA (r=0.9337,P<0.0001).The expression of HIF-1αmRNA in PCa was significantly related with the histological grading and clinical staging(P<0.05).The expression of HIF-1αmRNA is higher in poorly differentiated PCa than that in well and moderate differentiated PCa(P < 0.05).In addition,there was no significancant difference between HIF-1α,VEGF,GLUT-1 mRNA expression and tPSA (r1=0.4257,P=0.0782;r2=0.3514,P=0.1527;r3=0.4194,P=0.0832).Conclusion①The expression levels of HIF-1α,VEGF,GLUT-1 mRNA in PCa were higher than those in BPH.It's the reason why the expression levels of their proteins were high in PCa.②The high expression of HIF-1α,VEGF and GLUT-1 in prostate cancer tissues was significantly related with the histological grading and clinical staging. Detecting the level of their expression can be an important measure for diagnosis,staging and prognosis monitoring of PCa.③There are positive regulation between HIF-1αand VEGF gene. HIF-1αgene probably promotes tumor angiogenesis by regulating the transcription of VEGF gene.④There are positive regulation between HIF-1αand GLUT-1 gene. HIF-1αgene probably promotes tumor glucose transport by regulating the transcription of GLUT-1 gene.⑤There was a close correlation among HIF-1α,VEGF and GLUT-1 in PCa, which may play important roles during the development and invasion of PCa.Detecting the expression levels of them are practical for the diagnosis,staging and prognosis monitoring of PCa.
