Effects Of Androgen(Nandrolone Phenylpropionate)on T Cell Development Thymus And Spleen In Mice

The development of T cells differentiate and mature in the thymus,and then enter the peripheral immune system.As both thymus and spleen are important immune organs,they are closely linked with the aging and various diseases.Studies have confirmed that exogenous hormones can cause thymic atrophy of thymus gland,but for spleen effects are rarely reported.So which cells are effected by androgen in the thymus,and how does it work?To solve the above problems,we choose before and after puberty(1 weeks and 3 month old)Kunming mice as experimental subjects,and 3 concentrations of 10ug/10g,30ug/10g,50ug/10g of androgen(nandrolone phenylpropionate).Next intraperitoneal injection of 0.1ml for each time,we perform the continuous injection for 5 times every other day.The thymus and spleen tissue was weighed respectively.To observe the changes of structure of thymus cortex and medulla,the cell density changes of white pulp and red pulp in the spleen by paraffin section.Flow cytometry analysis was performed to compare changes of T cell subpopulations in the thymus and spleen.Through the above experiments,we get the following results:(1)Androgen(nandrolone phenylpropionate)activin may lead to a decrease of 1 week old mice and the weight of thymus and spleen index,causing atrophy in thymus and spleen tissues.(2)Comparing with different age mice,androgen can cause the atrophy of thymus at the age of both 1 weeks and 3 months,that long-term injection of exogenous androgen in childhood and adolescent mice have immunity inhibition effect.(3)As paraffin section observation showed that mice in 3 months,part of the thinning of thymus cortex,medulla increased,indicating that T cells mature in the thymus increased.Flow cytometry results showed that double positive T cells decreased,CD4 positive cells were slightly floating,which were consistent with the results of paraffin section.(4)Androgen has a stronger effect on prepubertal mice,which may be related to a lower physiological level of sex hormones in mice in one week.(5)The androgen nandrolone phenylproporonate exerted a stronger effect on the spleen of one-week-old Kunming mice than the thymus,but it was stronger on the thymus in the three-month-old male mice.(6)In the case of the same injection dose,there was a significant difference in thymus index and spleen index between the three-month-old female mice and male mice.According to the above results,we can draw the following conclusions:(1)Androgen(nandrolone phenylpropionate)will lead to different age mice atrophy of thymus.(2)Androgen mainly through the inhibition of double positive cells,lead to the atrophy of thymus.(3)Androgen have a stronger effect on male mice than female mice.(4)Androgen has different organizations preference in different age groups mice,in the other words,it effect on the different development stages of T lymphocytes.According to the above conclusion,we found that even a low dose exogenous androgen will down-regulate the immune function the of body,especially for prepubertal mice.So in clinical medicine,we need to strictly control the use of androgen in young patients.
