Purpose: Through the observation on the changes of IFN, IL-4, spleen index,thymus index and general state on rats with liver depression and spleendeficiency caused by electropuncture therapy, discussion of the effects ofelectroacupuncture on prevention of liver stagnation and spleen deficiency types,and observation of the changes of immune function, to study theelectroacupuncture preconditioning preventive mechanism, and to provideexperimental basis for prevention and treatment for the liver stagnation andspleen deficiency diseases, and to provide a better guide for clinical medicalcare application.Material and method:Select adult wistar rats-5male rats and5female ratsin each group. Based on their weight, randomly divide them into five groups:Control Group (no treatment), Model Group (standard treatment), Treatment Group1(" Taichong+Sanyinjiao”), Treatment Group2(" Taichong+Sanyinjiao+Tianshu"), Treatment Group3(" Taichong+Sanyinjiao+shangjuxu”). Seven daysbefore the experiment, treat Treatment Group1-3with electroacupuncturepreconditioning therapy for20minutes. During the8th-22nddays, except for GroupA, use chronic stress method (unpredictable) molding-methods include bound,no supply of water, fasting,45°C warm water swimming, and reversed day andnight. Arrange the schedule based on methods randomly drawn from the pool. Eachmethod is implemented three times, making a total of15-day modeling. When themodeling is done, observe the changes on rats’ weight and general conditions,test thymus index, spleen index, and IFN, IL-4protein expression level in therats’ colons. All the statistics are analyzed using the SPSS17.0.Results:1. General state: Control Group showed no significant abnormality. Compared withthe control group, model group showed apathy, depression, eating less, lyingwith tiredness, reduced fighting, loose stool thin, hair becoming dull, dry, frizzy and yellow. All the rats in Treatment Groups were less active, with noother obvious changes compared to control Group.2. Compared with control group, rats in the model group had no significant weightincrease (P <0.01). Treatment Groups had slight weight gain compared to modelgroup (P <0.05).3. Compared with control group, rats in the model group reduced spleen weightsignificantly (P <0.01). Rats in the treatment groups slightly increased theirspleen weight compared with the model group (P <0.05). No significant differencein thymus weight between each group (P>0.05),4. Compared to control group, the thymus index increased in the model group (P<0.01); Treatment groups showed decrease in thymus index compared with the modelgroup (P <0.05). No significant difference in spleen index between each group(P>0.05).5. Compared with control group, rats in the model group showed lower INF colonicexpression levels (P <0.01); Rats in the treatment groups showed higher colonicINF expression level compared with model group (P <0.05), no difference comparedwith control group (P>0.05)6. Compared with control group, rats in the model group showed colonic IL-4expression level (P <0.01). Treatment groups showed lower colonic IL-4expression level compared to model group (P <0.05), and no difference comparedwith control group (P>0.05)Conclusion:1. Acupuncture preconditioning is an effective way to prevent healthy rats from liver stagnation and spleen deficiency induced by chronic unpredictable stress.It is also an effective way in prevention of from liver stagnation and spleendeficiency related diseases.2. Through stimulation of corresponding acupoints, acupuncture preconditioningis able to regulate colonic INF and IL-4expression levels, effect thymus andspleen indexes, so as to improve the immune system. Conclusion might be drawnthat the acupuncture preconditioning mechanism realizes the prevention effectthrough adjusting and improving immune system in the bodies.3. Based on the experiment statistics, there is only small difference betweendifferent acupuncture methods. No evidence of different acupoints producedifferent expectations illustrates the difference of acupoints is mainlyreflected in the treatment but not prevention.
