Background:Primary liver cancer is a common malignant tumor of the digestive system.The current statistics show that the incidence and mortality of primary liver cancer in the world have been increasing year by year,and more than seven hundred thousand new cases of liver cancer are found in the world every year.Primary liver cancer is characterized by high degree of malignancy,early clinical atypical,rapid progress and poor prognosis.It has great threat to human health.The incidence of primary liver cancer in China is increasing year by year.The incidence of male disease is more than that of women.There are about two hundred and fifty thousand deaths per year in the world,of which the proportion of our country is about 40%.The mortality rate of this disease is third in digestive malignant tumor,second only to esophageal cancer and gastric cancer.Single nucleotide polymorphisms(Single nucleotide polymorphisms,SNPs)are polymorphic sequences of deoxyribonucleic acid caused by the variation of single nucleotide(G,A,C and T)at the gene level.A large number of studies have confirmed that single nucleotide polymorphisms account for ninety percent or more of the genetic variation in humans.In general,the change of a base,such as the transformation and transformation of a single base,can also be caused by deletion or insertion of bases.SNP not only has the function of genetic marker,but also influences the susceptibility to disease and prognosis of a patient by regulating gene expression.Related studies have shown that SNP in the coding gene region is associated with a variety of malignant tumors,including liver cancer.SNP is involved in a series of processes,such as the formation,progression and metastasis of liver cancer.G protein signal regulator(G protein signal regulator,RGS),which is combined with activated G alpha subunit,plays a negative role in regulating G protein signal.A member of the RGS family,a member of RGS5,is located in the chromosome 1q23 region,with a molecular weight of about 21kDa.It is located in the cytoplasm and membrane of cells in two different forms of monomers and two polymers.It is highly expressed in various tumor tissues and is of great significance to the reconstruction and maturation of tumor vessels.Objective:The purpose of this study was to analyze the correlation between the SNP of RGS5 and the survival rate after interventional therapy for the patients with primary liver cancer,and to provide a theoretical basis for finding the prognostic markers for the prognosis of primary liver cancer after interventional therapy.Methods:Select the secondary allele frequency(minor allele frequency,MAF)of the RGS5 gene region more than 10%SNP,and use PCR-RFLP technique to classify the blood samples of the patients after interventional therapy of 334 cases of liver cancer in Yanbian Korean Autonomous Prefecture of China.Analysis.Then,according to the complete clinical information of patients,SNP with statistical significance was screened out.Two genotyping pRL-SV40 plasmids were prepared and transfected into HCC cells,and SNP function experiments were carried out.Results:The analysis of the correlation between the RGS5 gene and the clinical data showed that only rs15049A>C in the 12 Poly-miRTS of the RGS gene had statistical significance in the disease free survival(P=0.03)and the total survival rate(P=0.06).The functional test results of SNP confirmed that the activity of rs15049A allele promoter was significantly higher than that of rs15049C allele,p= 0.003.Conclusion:1.The survival rate of rs15049AA allele in RGS5 3 'UTR was significantly lower than that of AC+CC,and the promoter activity of rs15049A allele was higher than that of rs15049C allele,which could be used as a marker for predicting survival rate in patients with primary liver cancer.2.The rs1504A>C of RGS5 gene is associated with the adverse effects of interventional therapy in patients with primary liver cancer.
