Study the chemical constituents and evaluate their in vitro anti-tumor activities of the flower Angelica acutiloba(Sieb.et Zucc)Kitagawa.The dried flowers of Angelica acutiloba(Sieb.et Zucc.)Kitagawa were extracted with 95%ethanol by heating reflux solvent,and the EtOH solution was partitioned with petroleum ether,EtOAc and n-BuOH,subsequently.The constituents were isolated by silica gel column chromatography,reversed-phase silica gel column chfomatography,HPLC,and the structures were identified by the spectral analyses,including 1H-NMR and 13C-NMR spectra,et al.The volatile components in dried flower of Angelica acutiloba were handled by gas purge microsyringe extraction(GP-MSE),and were analyzed by gas chromatograph-mass spectrometer(GC-MS).Area normalization method was used for the detection of the volatile component.The cell inhibition rates of monomeric compounds to HCT116,HT29,and A549 cells were measured by MTT assay.Sixteen compounds were finally obtained from the ethanol extract of the flower of Angelica acutiloba.and the structures were identified as apigenin(1),diosmetin(2),quercetin-3-O-glucopyranoside(3),quercetin-3-O-?-D-glucopyranosy-7-O-?-gen tiobioside(4),kaempferol-3-?-L-rhamnoside(5),kanferol-3?O-?-L-(4"-E-p-cumaroil)-raminoside(6),platanside(7),bergapten(8),E-6,7-tetrahydroxy-isobenzofuranone(9),senkyunolide H(10),senkyunolide I(11),protocatechuic acid(12),gallic acid(13),5-p-trans-Coumaroylquinic acid(14),1-dodecanol(15),and ?-sitosterol(16).Among these compounds,compounds 1,2,3,4,5,6,7 and 14 were firstly isolated from Angelica acutiloba(Sieb.et Zucc.)Kitagawa,and compounds 4,6,7 and 14 were isolated from this plant for the first time.In addition,the volatile components in the flower of East Angelica were examined.Among 77 components were identified,which accounting 51.88%of total contents.The content of hexadecanoic acid and 1(3H)-isobenzofuranone were the highest(5.28%)and 2,3-dihydrobenzofuran(4.36%).In vitro anti-tumor activities of the constitutes were determined by MTT assay,Compounds 6 and 7 showed significant inhibitory effects on HCT116 cells and HT29 cells(HCT116 cells IC50 ? 67.4 ?M,41.3 ?M,HT29 cells IC50 = 45.7 ?M,30.3?M),Cell growth has extremely significant significance,and the inhibitory intensity increases with increasing concentration.Compounds 6 and 7 are worthy of detailed advance research.
