B7-H4,a member of B7 family,is specifically expressed in many tumor tissues such as ovarian cancer,breast cancer,lung cancer,esophagus cancer,prostate cancer,gastric cancer,etc,and is associated with poor prognosis.The binding of B7-H4 with its unknown receptor induced negative immune regulation by inhibiting the proliferation and cytokine secretion of T cells,therefore producing tumor immune escape.Studies have shown that anti-tumor immune effect could be promoted by anti-B7-H4 antibody.Through the blockade of the binding of B7-H4 molecule to its receptor,the immune cells were activated to kill the tumor cells,suggesting that the role of B7-H4 receptor in tumor immunity is very important.Considering that B7-H4 receptor has not yet been cloned,the research about its receptor is rare,we have produced the monoclonal antibody to human B7-H4 receptor(10A10)by immunizing mice with tonsil cells expressing of B7-H4 receptor in previous study.In this study,10A10 antibody to B7-H4 receptor was used to explore the correlation between the expression of B7-H4 receptor in lung cancer tissue and the clinical parameters of lung cancer patients.First,the experiments were performed in lung cancer cell lines and lung cancer tissue slices to examine if the B7-H4 receptor was expressed,and then,lung cancer tissue microarray was bought and used to study B7-H4 receptor expression and the correlation with clinical parameters,so as to provide the new clue for clinical diagnosis and treatment of lung cancer.Part ?: 10A10 antibody to B7-H4 receptor was used to detect the expression of B7-H4 receptor in H1299 and A549,lung cancer tissues and adjacent tissues by immunohistochemistry.Objective: To detect the expression of B7-H4 receptor in paraffin sections of lung cancer and adjacent tissues and in lung cancer cell lines H1299 and A549.Methods: The expression of B7-H4 receptor in lung cancer cell lines H1299 and A549 was detected by flow cytometry using 10A10 antibody.The localization of B7-H4 receptor in cells was examined by cell staining.Immunohistochemistry was employed to detect the expression of B7-H4 receptor in lung cancer tissues and adjacent tissues.Results: The results from flow cytometry test showed that B7-H4 receptor was both expressed in lung cancer cell lines H1299 and A549,but the expression levels were different.B7-H4 receptor expression in A549 was slightly stronger than in H1299.The cell staining results also showed that the stronger staining could be seen in A549 cells than in H1299 cells,which was consistent with the results from flow cytometry test.In addition,the localization of B7-H4 receptor in H1299 and A549 existed mainly in the nucleus and cytoplasm.The immunohistochemistry results of lung cancer tissues and adjacent tissues displayed that B7-H4 receptor was stained mainly in nucleus and cytoplasm,and the coloration of cancer tissues was significantly stronger than that of adjacent tissues.Conclusion: B7-H4 receptor was expressed in lung cancer cell lines H1299 and A549 and lung cancer tissues,and existed mainly in the nucleus and cytoplasm.Part ?: Detection of B7-H4 receptor expression in lung adenocarcinoma tissue microarray using the antibody 10A10 and analysis of its correlation with clinical parameters.Objective: Detection of B7-H4 receptor expression in cancer tissues and adjacent tissues of patients with lung adenocarcinoma by 10A10 antibody and analysis of its correlation with clinical parameters.Methods: By immunohistochemistry,the expression of B7-H4 receptor in cancer tissues and adjacent tissues of 75 cases of patients with lung adenocarcinoma was detected by the anti-B7-H4 receptor 10A10 antibody.Score was obtained according to the staining intensity and positive rate of cells,and analysis of its correlation with clinical and pathological data was performed using SPSS statistics software.Results : The result showed that B7-H4 receptor was mainly expressed in cytoplasm and nucleus of lung adenocarcinoma,The expression level of B7-H4 receptor in lung adenocarcinoma tissues was higher than that in adjacent tissues.The correlation analysis showed that the expression of B7-H4 receptor in cancer tissue cytoplasm and adjacent tissue cytoplasm was positively correlated.The expression of B7-H4 receptor in cancer cell nucleus was also positively correlated with adjacent tissue nucleus.In addition,the expression of B7-H4 receptor in cancer cell nucleus was related to gender and clinical stage.But expression in nucleus of adjacent tissue is only related to the patient gender.Survival analysis showed that the expression of B7-H4 receptor in cancer tissues and in adjacent tissues is related to the prognosis of patients.In addition,tumor size,T stage,N stage and clinical stage were also significantly correlated with the prognosis of patients.In multivariate analysis,expression of B7-H4 receptor in nucleus of cancer cell and tumor size could be an independent prognostic factor for patient.Conclusion: The expression of B7-H4 receptor in lung adenocarcinoma is associated with the patient's prognosis,and its expression in the nucleus of cancer cell and tumor size could be an independent prognostic factor.
