| Object:To observe the effect of QufengHuoxue Pills on the expression of IL-17 mRNA,IL-23 mRNA and RORyt mRNA in the retina of experimental autoimmune uveitis(EAU)rats.Method:Eighty(160 eyes)male Lewis rats were randomly selected as blank group(A)according to the random number table method.The other 64 rats were injected subcutaneously with 0.1 ml of 30 μginter-photoreceptor retinoid binding protein(IRBP)1177-1191 between the hind legs.Freund’s complete adjuvant(FCA)was used once and Lewis rats were actively immunized to establish an EAU rat model.After modeling,the inflammation of the anterior segment of the eye was observed under a slit lamp microscope,and the inflammation score was performed according to the Caspi score.On the 7th day after immunization,the eyeballs of Lewis rats were used to make pathological sections and histopathological examinations were performed after HE staining.After the completion of the model,they were randomly divided into a model group(B),a regular dose group of QufengHuoxue Pill(C),a double dose group of QufengHuoxue Pill(D),and a XiongdanKaiming tablet group(E)according to the random number table method.After continuous intragastric administration for 14 days,real-time quantitative PCR(qPCR)was used to detect the relative expression of IL-17 mRNA,IL-23 mRNA and RORyt mRNA in rat retina tissue.Results:1.Validation of EAU rat model:On the 3rd day after immunization,the rats of the modeling group showed mild dilation of iris vessels,corneal vascular hyperemia,and ocular inflammation score(0.70±0.23)under a slit lamp microscope.On the 5th day,iris blood vessel dilation and congestion increased,anterior chamber opacity,corneal posterior wall deposit(KP),aqueous flare,miosis,and ocular inflammation score(2.60±0,42).On the 7th day,the iris blood vessels were significantly dilated and congested,the aqueous humor was turbid,the anterior chamber became shallow,the hysterospora,KP,the pupil margin gray floc,pupillary membrane closure,and the ocular inflammation score(3.10±1.21).On the 7th day after immunization,the pathological section of the eye of the rat showed various degrees of inflammatory cell infiltration in the posterior cornea,iris,ciliary body,and retina.The above shows that on the 7th day after immunization,the EAU rat model was successfully established.2.The results of qPCR showed that the relative expression of IL-17 mRNA was significantly higher in the model group than in the normal group(P<0.01).Compared with the model group,the QufengHuoxue Pill was used in the conventional dose group.The expression of XiongdanKaiming Tablets was decreased,and the difference was statistically significant(P<0.05).The expression of QufengHuoxue Pills in the double dose group was significantly lower(P<0.01).The relative expression of IL-23 mRNA:Compared with the normal group,the expression of the model group was significantly higher,the difference was statistically significant(P<0.01);Compared with the model group,the QufengHuoxue Pill double dose group,XiongdanKaiming film group The expression was decreased,and the difference was statistically significant(P<0.05).There was no significant decrease in the expression of QufengHuoxue Pills in the conventional dose group,and the difference was not statistically significant(P>0.05).The relative expression of RORyt mRNA:Compared with the normal group,the expression of the model group was significantly increased(P<0.01).Compared with the model group,the traditional dose group of QufengHuoxue Pills and the double dose group of QufengHuoxue Pills were compared.The expression of XiongdanKaiming Tablets was decreased,and the difference was statistically significant(P<0.05).Conclusion:1.Actively immunizing Lewis rats with IRBP antigen-adjuvant emulsifier successfully established an EAU rat model.2.QufengHuoxue Pill can inhibit the expression of IL-17 mRNA,IL-23 mRNA and RORyt mRNA in retinal tissue of EAU rats,indicating that IL-17,IL-23 and RORyt are the targets for the intervention of QufengHuoxue Pills in EAU.The purpose of improving the pathological process of EAU can be achieved by inhibiting the above targets.3.QufengHuoxue Pill may exert its anti-inflammatory effects by inhibiting the expression of Th17-associated inflammatory cytokines IL-17 mRNA,IL-23 mRNA and RORyt mRNA induced by uveitis. |
