| Objective: Preparation of allogeneic platelet-rich plasma and to observe the diabetic rats with deep II degree burns wound healing.Method: The SPF male SD rats(aged 8 weeks,weighing 200-250g)were randomly divided into 3 groups,Rats of group A and B were injected with streptozocin to induce diabetic models.Group C as normal control group,each group had 12 rats.Diabetic [60 mg/kg of STZ,single i.p.] group and normal [citrate buffer solution i.p.] group.72 h after injection with STZ,the blood glucose levels were monitored and compared form rat vena caudalis.Levels of blood glucose were evaluated at the 0th,1st,2nd,3rd,4th week respectively.The tissue sections of pancreas were stained by HE at the 8th week.After 8 weeks then type 1 diabetes rats after modeling successfully,using heated water bath 80℃,three group rats were produced deep II degree burn wound with a area is 4×4 cm2 was established on the back.Each grorp were used different interventions:The wound were treated with allogeneic platelet-rich plasma or N.S.dressing Daily dressing and measurement of wound area.Tissue samples from wound site were collected on in 7,14 and 21 d point were randomly killed four rats in each group take scalded skin full-thickness specimens for histological and immunohistochemical staining to detect the VEGF expression level,the microvessel density,inflammatory cells,fibroblasts and angiogenesis.Result: The blood glucose level reached the mold-guide standard and diabetic appearance occurred at 7 days after STZ administration.Compared with normal group,the pancreatic islets of diabetic rats were destroyed and shrunk,and the endocrine cells of pancreatic islets were markedly decreased in experiment rats.At the 14 th and 21 st days after scald,the wound healing rate of group A was significantly higher than that of group B(P<0.05).HE staining showed that the wounds of group B still had inflammatory cell infiltration on the 21 st day after scald.Compared with the diabetic control group,the MVD expression in the diabetic rats after PRP treatment for 7 days was significantly higher than that in the diabetic control group(P < 0.05).The expression of VEGF was lower in burned wounds of diabetic rats than in the normal at various time points(P<0.05).Conclusion: 1.Intraperitoneal through injection of STZ(60mg/kg)is an excellent method to induce type I diabetes in rats.2.Allogeneic platelet-rich plasma can accelerate the process of wound healing by improve the expression of VEGF,increases the microvessels density,and promotes proliferation of granulation tissue. |
