Abstract/Summary:
ObjectiveThis article aims to investigate the effect of allogeneic rat platelet-ric h plasma(PRP)gel on deep II° burn wound care in rats.MethodsForty-five male SD rats with healthy weight and weighing from 250 g to 300 g were randomly selected.Twenty experimental SD rats were randomly selected for abdominal aortic puncture,and allogeneic platelet-rich plasma was prepared by modified multiple centrifugation.And stored at-20℃ in aliquots,thawed and prepared into PRP gel at the time of use,the remaining 25 rats were simulated to construct a deep II° burn model,and 24 rats were randomly divided into 3 groups according to the random number table method.Eight groups(n=8)were established,and an animal model of deep II° burn wounds with a symmetrical diameter of 2cm in the rat’s dorsal spine was established.Two wounds per rat,and different nursing measures were given according to the experiment.Group A: allogeneic platelet-rich plasma(PRP)gel care group;Group B: MEBO moist burn ointment(MEBO)care group;Group C: negative control group,experimental animals in each group after burns were caged.The changes of granulation and healing of the wound tissue were observed at 24 h after the burn model and at the 5th,10 d,15d and 20 d of the wound care period.The residual wound diameter and the final complete healing time were measured,and the time was calculated.The wound area of the wound was randomly selected from each group of animals,and some burn wounds were cut out to prepare paraffin sections.HE staining and collagen-specific Masson staining were performed on the wound tissues to observe the collagen fiber proliferation and unit section.The number of neovascularization was determined.Finally,the expression of CTGF,VEGF and b-FGF protein in different groups were detected by Western blotting(WB).Results1.At the 5th,10 d,15d and 20 d time points,the wound healing area of the PRP group was larger than that of the MEBO group and the NC group,and the difference between the groups was extremely significant(p<0.01);the final healing time of the wound in the PRP group T(19.25±1.26d)was also significantly shorter than MEBO group(22.70±1.35d)and NC group(26.65±1.19d),the difference was significant(p<0.05);2.At the 5th,10 d,15d and 20 d time points,the wound healing and proliferation of PRP group and MEBO group were higher than those of NC group,and PRP group had the best effect;new capillary,hair follicle,sweat gland cells and catheter On the 5th and 10 th,the PRP group and the MEBO group grew faster than the NC group,and the PRP group had the best effect.On the 15 th and 20 th,the NC group increased significantly compared with the PRP group and the MEBO group;on the 5th and 10 d,The collagen content of each group showed an upward trend.The collagen content of PRP group and MEBO group was higher than that of NC group,and the PRP group was the highest,while the collagen content of each group showed a downward trend at 15 d and 20 d,and the expression of PRP group was significantly reduced.3.At the 5th day,the expressions of CTGF,VEGF and b-FGF in each group showed an upward trend,and the expression of PRP group was the highest,and the NC group was the least.The difference between the groups was extremely significant(p<0.01).At the 15 th day,the expression of CTGF protein in each group increased,but the increase rate decreased.The expression of VEGF and b-FGF protein decreased in each group,and the decrease in PRP group was the most obvious.The difference between groups was extremely significant(p<0.01).ConclusionPlatelet-rich plasma gel can effectively shorten the inflammatory phase reaction and reduce the exudation of wounds in the treatment of deep II° burns.In the period of granulation tissue proliferation,it can significantly increase the number of new capillaries,promote fibroblast proliferation,increase CTGF,VEGF,The expression of b-FGF protein effectively increases the collagen content,greatly shortens the final healing time,and greatly reduces the probability of infection and the probability of excessive granulation hyperplasia.
