bjective:To explore the role of the mechanism of burn wound healed using platelet rich plasma of rabbit,by establishing the scalding model, and applying of platelet rich plasma of rabbit, and observing burn wound healed. Methods:1. Making of platelet rich plasma gel of rabbit:before one day of the experiment, the two whole process of making prp was strict sterile which using landesberg law, and making the force by 200 g crystals.the prp gel would become jelly,mixed the prp,blood agent and little air in 6-10s.2. Twenty-four rabbits were randomly exposed to experimental group and control group (n=12 each).The mean weight were respectively (2.64±0.53) g, (2.73±0.52) g, not limited to male and female.Making the deep-Ⅱwound on the rabbit each side that was confirmed in the organization,which each side area was about 10cm and7cm,after put off the rabbit fur by vulcanization sodium,then used the hot water of 92℃burning the back of the rabbits.To fight the shock abdominal injected with saline water of 30ml/Kg after one hour of the experiment for the two groups.in six hour, the platelet rich plasma gel evenly spread to the wound for experimental group,which the thickness was about 1mm,binding up with 3m film and suturing their margins.the sulfadiazine silver paint to the wound for control group.3. Calculating the wound to heal rate that the wound to heal with the total area by the computer images and analysis of the fully automated system.4. In the experiment of 7,10,14 day, respectively, four rabbits which were randomly selected in the experimental group and the control group were killed after anesthesia.the skins would be removed and placed in 4% paraformaldehyde fixed,ethanol dehydrated progressively,embedded in paraffin sections and HE stained.Obverved the heal situation of skin by light mirror.5:Calculating and Contrasting a number of vessels for five view using microscope each group.6 The expression of bFGF,VEGF were observed in immunohistochemistry.The results of immunohistoche mistry would be semi-quantitative analysis using Motic digital medical image analysis system.7.All parameters were expressed as the mean value±standard difference(x±S),statistical analysis were carried out by the use of the least significant difference. Difference was considered significance when the p.value was less than 0.05. The statistics work were finished by spss17.0 software. Results:1.Afer the two methods treatment, the scalded area would reduce with time prolonged, especially the experimental group.On the seven day for the experimental group,the prp Firmly cemented in the margin and the wound oozes reduced. On the ten day, the wound oozes significantly reduced and the fur set grown significantly. On the fourteen day,the wound was dry with narrowed considerably,some which was soft and not keloids. But on the seven day for the control group, the wound oozes were up more,some which would appear sexual discharge pus. On the ten day, the wound oozes slightly reduced and some wound was further black. On the fourteen day,the wound was dry and some sulfadiazine silver was off.2. On the seven day,the wound appeared skin cells propagates,new capillaries and into the cells for the two group, especially the experimental group. On the ten day,the skin cells, new capillaries and into the cells were more for the two group. On the fourteen day, the new skin cells has been covered the wound basically for the experimental group,but the proliferation cells are more active for the control group.3. a number of vessels for five view using microscope each group was Obvious statistics differences.4. the wound to heal rate was obvious statistics differences for the two group.5, the cells or (and) the nucleus of brown were positive cells by the mean of Immune organization chemical. A quantitative work for bFGF and VEGF.On 7,10d,the expression of bFGF and VEGF were both higher and obvious statistics differences for the two group (P<0.05),but on 14d,which gradually declined and was not obvious statistics differences for the two group(P>0.05). Conclusion The main mechanism of platelet rich plasma bringing healing burns big is promoting wound into a fabric with alkaline cell and the generated,and then The wound healed.
