Objective:To observe the effect of Shenling Baizhu Powder on the levels of IL-13,IL-23 and COX-2 in the colon of UC rats with spleen deficiency syndrome,and to explore the possible mechanism of action of Shenling Baizhu Powder on UC with spleen deficiency syndrome from inflammatory reaction,gene and protein.Methods:Sixty SPF Wistar rats were randomly divided into blank group and model reconstruction group according to body weight.Model rehabilitation group using compound method for 3 weeks to build UC rats with spleen deficiency syndrome model,DAI score,observe the colon mucosa and other methods to assess the success of modeling,model reconstruction group according to body weight and male and female were divided into model group,Shenling Baizhu powder?high,medium and low?and Mesalazine group.The blank group and model group were gavaged with distilled water 10 m L·kg-1·d-1,Shenling Baizhu Powder high?medium and low dose group according to the corresponding concentration of drug decoction(24?12?6g·kg-1·d-1)by gavage,mesalazine group intragastric administration of mesalazine enteric-coated tablets 0.2g·kg-1·d-1,all above continuous intervention for 21days.The contents of IL-13 and IL-23 in serum of rats were detected by ELISA,the expressions of IL-13,IL-23 and COX-2 mRNA were detected by RT-qPCR and the expression of COX-2 Protein expression were detected by WB.Results:1.Compared with the blank group,the colon tissues of the model group and each intervention group showed different degrees of infiltration,congestion and edema under light microscope.Compared with the model group,the scores of colonic mucosal injury of rats in each intervention group were decreased,especially in the Shenling Baizhu Powder high dose group?P<0.05?.2.DAI score:Compared with the blank group,DAI scores in model group increased significantly,the difference was statistically significant?P<0.05?;Compared with the model group,Except for the Shenling Baizhu Powder low dose group,the DAI scores of the other intervention groups decreased,the difference was statistically significant?P<0.05?;There was no significant difference in DAI score between Shenling Baizhu Powder high dose group and mesalamine group.3.Serum levels of IL-13 and IL-23 in each group were detected by ELISA:compared with the blank group,the content of IL-13 in the model group was significantly lower?P<0.05?and the content of IL-23 was higher?P<0.05?.Compared with the model group,the content of IL-13 in the intervention group was significantly?P<0.05?,especially in the Shenling Baizhu Powder high dose group and Mesalazine group,the difference was statistically significant?P<0.05?;The serum levels of IL-13 and IL-23 in the Shenling Baizhu Powder high dose group were not significantly different from that of mesalamine group?P>0.05?.4.The mRNA expressions of IL-13,IL-23 and COX-2 in the colon of rats in each group were detected by RT-qPCR:compared with the blank group,the expression of IL-13 mRNA in the colon decreased?P<0.05?and the expression of IL-23 and COX-2 m RNA in the model group increased?P<0.05?.Compared with model group,the expressions of IL-13 mRNA in the colon of Shenling Baizhu Powder group were higher than those in the model group,especially in the Shenling Baizhu Powder high dose group?P<0.05?,IL-23?COX-2 mRNA expression decreased?P<0.05?;Compared with the mesalazine group,the expression levels of IL-13,IL-23,and COX-2 mRNA in the colon of Shenling Baizhu Powder high dose group increased slightly,and the difference was statistically significant?P>0.05?.5.WB method was used to detect the relative expression of COX-2 protein in colonic tissue in each group:compared with the blank group,the relative expression of COX-2 protein in the colon tissue of the model group rats increased?P<0.05?;compared with the model group,the relative expression of COX-2 protein in the colon tissues decreased,Shenling Baizhu Powder high dose group was particularly significant?P<0.05?;Compared with the mesalazine group,the relative expression of COX-2 protein in the colon of Shenling Baizhu Powder high dose group was slightly higher,and the difference was statistically significant?P>0.05?.Conclusion:1.Shenling Baizhu Powder can improve the general survival of UC rats with spleen deficiency syndrome,reduce the damage of colon tissue,especially in Shenling Baizhu Powder high dose group.2.IL-13,IL-23,COX-2 in UC rat with spleen deficiency syndrome serum or colon reflect,and the severity of the disease has a certain guiding significance.3.Shenling Baizhu Powder can improve the quality of life and inhibit the inflammatory reaction in rats with spleen deficiency syndrome,which may be related to the increase of IL-13 and the decrease of the expression of IL-23 and COX-2.
