Chuanxiong is derived from the dried rhizomes of the Ligusticum chuanxiong Hort.It is famous for medicinal hebs of Sichuan province.It is mild and flavorful,and is infused with liver and heart.It has the effect of invigorating blood and removing wind and relieving pain.It is commonly used to treat headache,and it is also praised as "blood gas medicine".The decoction pieces of Chuanxiong Rhizoma and prepared products are commonly used,which has a wide medicinal value.However,there is a lack of unified production technology standard and scientific organization management,which leads to the uneven quality of chuanxiong slices.Therefore,it is necessary to establish a comprehensive quality evaluation system for the standardized study of chuanxiong Rhizoma,in order to better guide the clinical medication and the standardized production of decoction pieces of chuanxiong Rhizoma.This article mainly study from the processing technology,content determination and fingerprint,the respect such as preliminary pharmacodynamic evaluation,further perfecting and improving their quality standards,for standardization of decoction pieces of chuanxiong Rhizoma provides a theoretical basis.The main research contents are as follows: 1.The processing technology of decoction pieces of chuanxiong Rhizoma was studied from the "net-softening-cutting-drying".The optimized process is: removing the fibrous root,silt and impurities from chuanxiong Rhizoma,add the 20% of the weight water,and make 24 h,cut into 2~4mm thick pieces,and dry 5h in oven.A multi-index comprehensive weighted scoring method was used to combine the processing technique of the optimal design response surface method.Firstly,single factor test was used to investigate the dilution and time of the yellow wine.Then,the important parameters in the processing of chuanxiong chuanxiong were added,and the temperature and time of cooking were independent variables,and D-optimal design was carried out.The OD values of the five active ingredients,ferulic acid,lignin,ligustron A,ligustrum,and 3-n-butylphenylphthalein,were used as dependent variables.Through the analysis of the response surface,the processing technology of the optimized chuanxiong slices was obtained.Adding 20% rice-wine,mixed with 20% the quality of rice wine with diluted,mixed with decoction pieces of chuanxiong Rhizoma,stuffy embellish 75 min,and making 12 min at 156℃.2.A method was established for the determination of the five active components of ferulic acid,ligusticate,ligusticate,ligusticin,and 3-n-butylphenylphthalein.The content control method is good in precision,stability and repeatability,and can be used as an effective component of chuanxiong.The fingerprint analysis of decoction pieces of chuanxiong Rhizoma and prepared products was carried out by high performance liquid chromatography,and similarity evaluation was carried out by using Chinese medicine fingerprint similarity evaluation software.Among them,11 were confirmed,and 5 characteristic peaks were identified.The characteristic fingerprint was established.3.The preliminary pharmacodynamics evaluation of chuanxiong slices and their alcoholic products of different grades were carried out.Analgesic experiment(hot plate method,torsion method)was used to simulate the hemorheology of rat acute blood stasis model.To study the analgesic and activating blood circulation in different grades of chuanxiong yinpian and its wine.The results showed that different grades of chuanxiong chuanxiong and its alcoholic products could increase the pain threshold of the mice.All of them can prolong the incubation period and reduce the number of torsion in mice induced by acetic acid.It can reduce the total blood shear rate,plasma viscosity,erythrocyte aggregation index.The analgesic,the blood circulation of the first class is better than the second and third.At the same dose,the pharmacokinetics of the wine was more significant than the product,which was consistent with the theory that "alcohol can lead to ascending and enhance the action of activating blood and relieving pain".This article mainly from the above three aspects to carry out the processing technology,content determination and fingerprint,preliminary pharmacodynamics research.
