Objective:Do research on the effects of Non-steroidal anti-inflammatory drugs?NSAIDs?on the level of Prostaglandin E2?PGE2?and Prostaglandin I2?PGI2?of patients with upper gastrointestinal bleeding by testing the level of PGE2 and PGI2 of patients with and without upper gastrointestinal bleeding after taking NSAIDs and patients with that effect but no taking NSAIDs,while discovering the effects of Helicobacter pylori?Hp?on both above according to the existence of Hp infection.Methods:To collect 35,12,and 33 cases of the 3 types of patients concerned respectively in our hospital from May 2017 to January 2018.The study subjects required no history of major organ failure and malignancy,no taking of prostaglandin-stimulating drugs?such as teprenone,gefitafil,etc.?in the past 6 months,no use of acid inhibitors in the past 3months,no long-term drinking history,cirrhosis,or esophageal and gastric varices,and no recent stress events like surgery,trauma etc.Those who have not taken NSAIDs additionally require that no other anticoagulant drugs be used.Blood samples of fasting elbow veins were collected from all subjects in the morning,centrifuged,and frozen at-70?ultra-low temperature refrigerator for testing.At the same time,general information such as the number,age,gender,type of medication,and dosage of medications of all subjects were recorded.Levels of serum PGE2 and 6-keto-PGF1?of subjects were measured by enzyme-linked immunosorbent assay?ELISA?.Results:.1.The levels of serum PGE2 and 6-keto-PGF1?of three groups concerned:The levels of PGE2 and 6-keto-PGF1?in the non-bleeding group were significantly higher than those in the medication-bleeding group and the untreated group.There was a statistical difference among the three groups?P<0.05?.The levels of PGE2 and6-keto-PGF1?in the medication-bleeding group and the untreated group were lower than those in the non-bleeding group.The differences was of statistical significance?P<0.05?.2.Comparison of serum PGE2 levels in the three groups of Hp-negative patients:the levels of the non-bleeding group were higher than those in the medication-bleeding group and the untreated group.There was a statistically significant difference among three groups?P<0.05?.Compared with the non-bleeding group,the levels of serum PGE2 in medication-bleeding group and the untreated group was decreased,and the difference was of statistical significance?P<0.05?.3.Comparison of levels of serum PGE2 in three groups of Hp-positive patients:the levels of medication-bleeding group were significantly higher than those in the non-bleeding group and the untreated group.There was significant difference among three groups?P<0.05?.Comparison of levels between no bleeding group and untreated group was of statistically significance?P<0.05?.The serum PGE2 levels in Hp-negative patients who received medication without bleeding were significantly higher than those with Hp-positive patients?P<0.05?.4.Comparison of serum 6-keto-PGF1?levels in the three groups of Hp-negative patients:The difference was of statistical significance?P<0.05?.Compared with the non-bleeding group the level of serum 6-keto-PGF1?in medication-bleeding group and the untreated group was decreased,and the difference was of statistical significance?P<0.05?.The serum 6-keto-PGF1?levels in Hp-negative patients who received medication without bleeding were significantly higher than those with Hp-positive patients?P<0.05?.Conclusion:1.The levels of serum PGE2 and 6-keto-PGF1?in the non-bleeding group after taking NSAIDs were higher than those in the bleeding group after taking NSAIDs and the non-NSAIDs bleeding group.2.In the bleeding group taking no NSAIDs,Hp has no effect on PGE2,6-keto-PGF1?levels.3.In the non-bleeding group taking NSAIDs,the levels of serum PGE2 and6-keto-PGF1?in the Hp-positive group were lower than those in the Hp-negative group,suggesting that Hp infection may synergize with NSAIDs.
