Objective:To study the effect of zinc ions on bicalutamide inhibited the proliferation of prostate cancer PC-3 cells,and try to explore its possible mechanism.Methods:The PC-3 cells were divided into four groups: the blank control group;10?M bicalutamide group;200?M zinc group;200?M zinc combined with 10?M bicalutamide group.To observe the effect of zinc ions combined with bicalutamide on cell morphology through the microscope;the cell survival rate in each group was determined by CCK-8 method;Cell apoptosis and cell cycles were detected by flow cytometry;The mRNA expression of matrix metalloproteinase-2(mmp-2),matrix metalloproteinase-9(mmp-9),cysteinyl aspartate-specific proteinase-3(Caspase-3)and cysteinyl aspartate-specific proteinase-9(Caspase-9)in each group were detected by Real-Time quantitative PCR(RTPCR).Results:1.Survival rate of PC-3 cellsCompared with the blank control group,bicalutamide group and znic group,the survival rate of PC-3 cell in zinc combined with bicalutamide group was significantly reduced(P<0.05).It showed that the zinc ions combined with bicalutamide can further inhibit the proliferation of PC-3 cells.2.Apoptosis rate and cell cycle of PC-3 cellsThe results showed that the apoptosis rate of zinc combined with bicalutamide group was significantly higher than that of control group,bicalutamide group and Zn group(P < 0.05),the G2/M phase was obviously blocked(P < 0.05).It indicated that zinc ions combined with bicalutamide can significantly promote the apoptosis of PC-3 cells.3.Expression of related genes of PC-3 cellsThe figures illuminated that,compared with the control group,bicalutamide group,zinc group,zinc combined with bicalutamide group can significantly reduce the mRNA expression level of mmp-2 and mmp-9(P < 0.05),and more significantly enhanced the mRNA expression level of Caspase-3 and Caspase-9 in PC-3 cells(P < 0.05).Conclusion:Zinc ions combined with Bicalutamide can remarkably inhibit the proliferation of prostate cancer PC-3 cell and promote cell apoptosis.The mechanism may be to reduce the gene expression of mmp-2 and mmp-3,and enhance the gene expression of Caspase-3 and Caspase-9 in PC-3 cells of prostate cancer.In some ways,Zinc ions enhanced the effect of androgen deprivation on prostate cancer PC-3 cells.Therefore,Zinc supplements may be better in the treatment of prostate cancer during the traditional endocrine therapy.
