Objective:To study the effect of radiofrequency ablation(RFCA)on the expression of c-Met protein in the rabbit model of VX2 hepatocellular carcinoma(HCC).Methods:About 1 years a total of 20 healthy New Zealand white rabbits were randomly selected as two tumor passaged rabbit VX2 tumor cell culture solution,will be planted in the passage of rabbit hind lateral muscle,the remaining 18 were used as experimental rabbits under anesthesia VX2 tumor implant was built in the left lobe of liver of rabbit rabbit liver tumor model,ultrasonic continuous monitoring,built rabbit tumor model.They were randomly divided into 3 groups(group A,group B,group C),6in each group,group A was tumor model control group,group B was single radiofrequency ablation group,C group was multiple radiofrequency ablation group(three times).Radiofrequency ablation at the same time after ultrasound examination,anesthetized three groups of rabbits,tumor body,increased anesthetic to the experimental rabbits in euthanasia.The expression of c-Met protein in all specimens was obtained by immunohistochemistry.Analysis of statistical methods.Results:18 rabbits were successfully implanted with VX2 tumor after 4W implantation.The round or round hypoechoic masses of liver around 1.5cm were observed under ultrasound.B group after radiofrequency ablation: the experimental rabbit tumor body ultrasound showed irregular shape,and the boundary was not clear.The center of 4W treatment area was irregular,there was no significant blood flow signal,no echo necrosis area,residual tumor area increased to preoperative,no capsule,no obvious blood flow signal.Group C: the experimental rabbit tumor body ultrasound showed irregular shape,and the boundary was not clear.After three radiofrequency ablation,there was irregular echo in the 4W treatment center,irregular echo around the center,and the tumor in the residual cancer area increased to 1.5 times than that before operation.HE staining group A: large deep staining of the nucleus of the tumor,nests distribution,large heteromorphism.Group B,group C: large deep staining,nestlike distribution and large heteromorphism in the tumor body of the residual cancer area.There was obvious cell necrosis in the treatment area,a large number of red dye tissues and more lymphocytes.Conclusion : After a period of radiofrequency ablation,the tumor can promote the expression of c-Met protein through some specific ways,resulting in stronger replication and invasion ability of tumor cells.
